Active Controlled and Tunable Coacervation Using Side-Chain Functional α-Helical Homopolypeptides

Summary: Developed new side-chain amino acid-functionalized alpha-helical homopolypeptides can reversibly form coacervate phases in aqueous media. Their multifunctional side-chains can actively control coacervation through mild, biomimetic redox chemistry and respond to changes in pH, temperature, and counterions, mimicking properties observed in natural coacervate forming intrinsically disordered proteins. Despite alpha-helical conformations that traditionally disfavor coacervation, molecular dynamics simulations suggest that high side-chain conformational disorder and hydration around the ordered backbone may explain the ability of these polypeptides to form coacervates.

Understanding How Coacervates Drive Reversible Small Molecule Reactions to Promote Molecular Complexity

Summary: This study demonstrates that coacervate droplets promote higher concentrations of reactants and products compared to bulk solution, due to preferential partitioning and higher reaction equilibrium constant in the droplets. A reaction-diffusion model reveals how competing reaction and partitioning equilibria govern the spatial distribution of products inside coacervate droplets. These results offer insights into how biomolecular condensates stabilize labile reaction products and promote molecular complexity through multistep reaction pathways involving unstable intermediates.
