Design, synthesis, biomedical investigation, DFT calculation and molecular docking of novel Ru(II)-mixed ligand complexes

Sara E. Mohamed et al.

Summary: A series of bioactive water-soluble mononuclear Ru(II)-mixed ligand complexes, containing 2,2'-bipyridyl and V-shaped Schiff base ligands, were synthesized and characterized. Their biomedical activities, including antioxidant activities, interaction with CT-DNA, and anticancer performance, were investigated. The optimized structure of these complexes was supported by DFT calculations, and molecular docking studies with DNA using MOE software further validated the interaction of some complexes.

Substrate stiffening promotes VEGF-A functions via the PI3K/Akt/mTOR pathway

Amjad Husain et al.

Summary: The study reveals the underlying role of the PI3K/Akt/mTOR signaling pathway in regulating vascular endothelial growth factor-A (VEGF-A) mediated signaling and functions. The pathway is activated on stiffer substrates, amplified by VEGF-A stimulation, and correlated with enhanced endothelial cell (EC) proliferation, contraction, pro-angiogenic secretion, and capillary-like tube formation. These findings have important implications for understanding age-related macular degeneration and provide insight into potential therapeutic pathways.

Summary: The synthesis and characterization of two ruthenium (II) polypyridyl complexes were reported, wherein it was found that these complexes bind to DNA via a minor groove mode of binding and can induce apoptosis of MCF-7 cells at low concentrations. Additionally, confocal microscopy showed that one of the complexes localizes in the nucleus and is more efficient at internalizing in MCF-7 cells than in HEK-293 cells.
