Contribution of senescent and reactive astrocytes on central nervous system inflammaging

Michel Lopez-Teros et al.

Summary: Astrocytes in the aging central nervous system have both neuroprotective and neurotoxic functions, contributing to chronic inflammation and impaired cognitive function. The secretomes of senescent and reactive astrocytes have not been directly compared in the aging brain.

A Monoclonal Anti-HMGB1 Antibody Attenuates Neurodegeneration in an Experimental Animal Model of Glaucoma

Henrik Tonner et al.

Summary: In this study, intravitreal injection of anti-HMGB1 Ab was found to reduce HMGB1-dependent inflammatory signaling and improve RGC neuronal survival in an experimental animal model of glaucoma. Mass spectrometric analysis demonstrated increased abundance of RNA metabolism-associated hnRNPs in animals injected with the antibody, while microarray analysis showed a decreased expression of inflammatory mediators in the HMGB1 group. These findings suggest that anti-HMGB1 Ab may play a neuroprotective role in glaucoma by modulating inflammatory processes.

Blood-Brain Barrier Dysfunction and Astrocyte Senescence as Reciprocal Drivers of Neuropathology in Aging

Marcela K. Preininger et al.

Summary: As the most abundant cell type in the brain, astrocytes play essential roles in maintaining brain homeostasis, regulating immune response, and protecting the blood-brain barrier. However, dysfunction of astrocytes is closely associated with age-related neuropathology and neurodegenerative diseases. This review discusses two mechanisms by which astrocytes mediate neuropathology in the aging brain – age-associated blood-brain barrier dysfunction and senescence-induced neuroinflammation. Understanding the relationship between these mechanisms and astrocytes is crucial for exploring the aging process and neurodegenerative diseases.

The Multifaceted Neurotoxicity of Astrocytes in Ageing and Age-Related Neurodegenerative Diseases: A Translational Perspective

David S. Bouvier et al.

Summary: Astrocytes play multiple roles in the central nervous system, but in age-related neurodegenerative diseases, they may exhibit neurotoxic phenotypes that contribute to disease progression. Chronic inflammation, oxidative stress, impaired phagocytosis, and altered physiological roles are the main manifestations of astrocytic toxicity. However, the involvement of astrocytes in brain deterioration in patients with neurodegenerative diseases is still controversial due to overlapping pathologies and technical challenges in studying human brain samples. This review provides an overview of astrocyte neurotoxicity from in vitro findings to animal models and patient-related research, and discusses the role of aging in astrocytes and potential therapeutic strategies.

Regionally encoded functional heterogeneity of astrocytes in health and disease: A perspective

Benjamin E. Clarke et al.

Summary: Increasing evidence suggests that astrocytes exhibit functional heterogeneity in different brain regions, with their diversity determining a spectrum of physiological states. They are now seen as viable targets for therapies aiming to restore neural circuit integrity in neurodegenerative disorders, and understanding their physiology and functional diversity will guide therapeutic strategies in these diseases.

Restoring metabolism of myeloid cells reverses cognitive decline in ageing

Paras S. Minhas et al.

Summary: Ageing is characterized by chronic pro-inflammatory responses that affect various aspects of health, including the brain. The study shows that in ageing mice, myeloid cell bioenergetics are suppressed in response to increased signalling by the lipid messenger prostaglandin E-2 (PGE(2)), leading to maladaptive pro-inflammatory responses. By inhibiting myeloid EP2 signalling, it is possible to reverse cognitive ageing and restore youthful immune functions.

Targeting Diet and Exercise for Neuroprotection and Neurorecovery in Glaucoma

James R. Tribble et al.

Summary: Glaucoma, a leading cause of blindness, involves dysfunction and death of retinal ganglion cells, with current therapies focusing on lowering intraocular pressure. However, emerging evidence suggests metabolic deficiencies may play a key role in glaucoma pathophysiology. Modifying diet and exercise to maintain metabolic fitness could provide therapeutic benefits to retinal ganglion cells in glaucoma.
