High-speed fluorescence image-enabled cell sorting

Daniel Schraivogel et al.

Summary: In this study, a high-speed image-enabled cell sorting method was established to isolate single cells with unique spatial and morphological traits. This method allows for quantitative analysis of cell morphology and localization of labeled proteins, and improves the resolution of cell cycle analysis. By combining it with CRISPR-pooled screens, comprehensive genome-wide image-based screens can be completed in a short period of time.

Live-seq enables temporal transcriptomic recording of single cells

Wanze Chen et al.

Summary: Live-seq is a novel single-cell transcriptomic profiling approach that preserves cell viability during RNA extraction, allowing for the correlation analysis between a cell's ground-state transcriptome and its downstream molecular or phenotypic behavior. It accurately stratifies diverse cell types and states without major cellular perturbations, and can be used to map a cell's trajectory and evaluate gene effects on cell phenotypes.

Proteome-wide mapping of short-lived proteins in human cells

Jiaming Li et al.

Summary: By combining cycloheximide chase assays with advanced quantitative proteomics, this study mapped short-lived proteins under translational inhibition in four human cell lines. Short-lived proteins are less abundant, evolutionarily younger, and less thermally stable compared to other proteins, with 103 proteins showing different stabilities among cell lines. Truncated forms of certain proteins in U2OS and HCT116 cells were also identified, indicating lower stability than full-length counterparts. This research provides a large-scale resource for understanding human short-lived proteins and their potential for therapeutic interventions.

Six distinct NFκB signaling codons convey discrete information to distinguish stimuli and enable appropriate macrophage responses

Adewunmi Adelaja et al.

Summary: The dynamics of NF kappa B signaling can classify immune threats through six signaling codons, and confusion of these codons may underlie the etiology of some inflammatory diseases.NF kappa B activity determines the type of gene expression and response in macrophages. An oscillatory trajectory is a hallmark of responses to cytokine stimuli.
