Distribution-Free Multisample Tests Based on Optimal Matchings With Applications to Single Cell Genomics

Somabha Mukherjee et al.

Summary: In this article, a nonparametric graphical test based on optimal matching is proposed to assess the equality of multiple unknown multivariate probability distributions. The test has been proven to be universally consistent and demonstrated superior performance through simulation studies. It has also shown meaningful results when applied to single cell transcriptomics data.

Defective STAT5 Activation and Aberrant Expression of BCL6 in Naive CD4 T Cells Enhances Follicular Th Cell-like Differentiation in Patients with Granulomatosis with Polyangiitis

Sangmi Kim et al.

Summary: Granulomatosis with polyangiitis (GPA) is a potentially fatal small vessel vasculitis characterized by antineutrophil cytoplasmic autoantibodies, chronic inflammation, and granulomatous tissue damage. This study found that dysregulation of T cell regulation and aberrant expression of naive CD4 T cells are closely associated with disease pathogenesis in GPA.

Arachidonic acid-regulated calcium signaling in T cells from patients with rheumatoid arthritis promotes synovial inflammation

Zhongde Ye et al.

Summary: In this study, it was shown that CD4(+) T cells from patients with RA and PsA have increased expression of the pore-forming calcium channel component ORAI3 due to reduced IKAROS expression, leading to increased ARC channel activity and sensitivity to arachidonic acid. This heightened activity may contribute to chronic inflammation in these conditions, indicating a potential mechanism for the pathogenesis of RA and PsA.

Integrated analysis of multimodal single-cell data

Yuhan Hao et al.

Summary: The study introduces a weighted-nearest neighbor analysis framework to learn the relative utility of each data type in each cell, enabling integrative analysis of multiple modalities. Applied to a CITE-seq dataset, the method constructs a multimodal reference atlas of the circulating immune system and successfully identifies and validates previously unreported lymphoid subpopulations.

UCell: Robust and scalable single-cell gene signature scoring

Massimo Andreatta et al.

Summary: UCell is an R package designed for evaluating gene signatures in single-cell datasets. It offers robust signature scores based on the Mann-Whitney U statistic, requiring less computing time and memory compared to other methods. UCell can handle large datasets in minutes and can interact directly with Seurat objects.
