Adenosine triphosphate binding cassette subfamily C member 1 (ABCC1) overexpression reduces APP processing and increases alpha- versus beta-secretase activity, in vitro

Wayne M. Jepsen et al.

Summary: ABCC1 has been shown to export Abeta from human cells while reducing its production, and promote the cleavage of APP by alpha- versus beta-secretase, possibly through modulation of secretase activity.

Deletion of Alzheimer's disease-associated CD33 results in an inflammatory human microglia phenotype

Jannis Wissfeld et al.

Summary: Genome-wide association studies have shown that mutations in the CD33 gene are linked to late-onset Alzheimer's disease. Deletion or Exon 2 deletion expression of CD33 in macrophages and microglia led to increased inflammatory pathways and activation of cytokines. The knockout of CD33 also resulted in enhanced phagocytosis of amyloid beta and bacterial particles, but potentially detrimental oxidative burst and inflammation.

Efficient manipulation of gene dosage in human iPSCs using CRISPR/Cas9 nickases

Tao Ye et al.

Summary: Researchers demonstrate a simple and precise method to simultaneously generate iPSC lines with different gene dosages using paired Cas9 nickases. As proof-of-concept they apply this method to examining amyloid precursor protein gene dosage effects in an Alzheimer's disease patient-derived iPSC line. Their method could potentially advance what we know about disease mechanisms and assist with gene therapy development.
