Selective Single-Cell Expansion on a Microfluidic Chip for Studying Heterogeneity of Glioma Stem Cells

Peiwen Li et al.

Summary: The study reveals the heterogeneity of GSCs in glioma and its correlation with morphological differences. Using a microfluidic single-cell culture approach, the researchers successfully expanded GSCs and classified them into three groups based on their morphological characteristics. Functional and transcriptomic analysis showed differences among these subtypes.

Phenotypic and molecular states of IDH1 mutation-induced CD24-positive glioma stem-like cells

Sara Haddock et al.

Summary: Mutations in IDH1 and IDH2 drive the development of gliomas by inducing phenotypic reprogramming and stem-like properties. The CD24-positive cell population in IDH-mutant tumors possesses remarkable stem-like characteristics and shows enhanced tumor initiating potency. The induction of a CD24-positive population is one mechanism by which IDH-mutant tumors acquire stem-like properties.

Differentiation therapy of myeloid leukemia: four decades of development

Vikas Madan et al.

Summary: Differentiation therapy in the treatment of acute myeloid leukemia is gaining more attention, with agents inducing cell differentiation acting on various cellular processes including dysregulated metabolic programs, signaling pathways, epigenetic machinery, and the cell cycle. Inhibitors of mutant IDH1/2 and FLT3 have shown clinical benefit and gained approval for use.

Integrated analysis of multimodal single-cell data

Yuhan Hao et al.

Summary: The study introduces a weighted-nearest neighbor analysis framework to learn the relative utility of each data type in each cell, enabling integrative analysis of multiple modalities. Applied to a CITE-seq dataset, the method constructs a multimodal reference atlas of the circulating immune system and successfully identifies and validates previously unreported lymphoid subpopulations.

Neural G0: a quiescent-like state found in neuroepithelial-derived cells and glioma

Samantha A. O'Connor et al.

Summary: Single-cell RNA sequencing was used to examine the cell cycle states of expanding human neural stem cells (hNSCs), revealing a quiescent-like state in neuroepithelial-derived cells and gliomas. Neural G0 markers were enriched with quiescent NSC genes, and were significantly associated with less aggressive tumors and extended patient survival in gliomas. Knockout of genes associated with the Hippo/Yap and p53 pathways diminished Neural G0, resulting in faster G1 transit and down-regulation of quiescence-associated markers.
