Metabolic determination of cell fate through selective inheritance of mitochondria

Julia Dohla et al.

Summary: The study demonstrates that selectively and asymmetrically inherited mitochondria in mammary stem-like cells impose a metabolic bias on progeny, affecting the balance between self-renewal and differentiation. This metabolic feature, established by functionally distinct mitochondrial age-classes, acts as a fate determinant in asymmetric division of epithelial stem-like cells.Cells that inherit old mitochondria with oxidative energy metabolism promote differentiation, while cells inheriting newly synthesized mitochondria with low levels of Rieske iron-sulfur polypeptide 1 maintain stemness through the pentose phosphate pathway during early fate determination.

Bioenergetic Metabolism In Osteoblast Differentiation

Leyao Shen et al.

Summary: Bioenergetic metabolism plays an important regulatory role in osteoblasts. Recent studies have identified the major nutrients and bioenergetic pathways utilized by osteoblasts, as well as their regulation during differentiation. This review highlights the acquisition and metabolism of glucose, amino acids, and fatty acids by osteoblasts to provide energy and metabolic intermediates, and discusses the signals that regulate nutrient uptake and metabolism, emphasizing how energetic metabolism promotes osteoblast differentiation.

Vitamin D and Its Target Genes

Carsten Carlberg

Summary: The vitamin D metabolite 1 alpha,25-dihydroxyvitamin D-3 acts as a ligand for the transcription factor vitamin D receptor (VDR), and regulates the expression of target genes through chromatin changes. VDR binds to genomic DNA and interacts with partner proteins.

Bone marrow and periosteal skeletal stem/progenitor cells make distinct contributions to bone maintenance and repair

Elise C. Jeffery et al.

Summary: The contribution of skeletal stem/progenitor cells (SSCs) from bone marrow and periosteum to bone repair has been investigated. During steady-state conditions, new bone primarily arises from bone marrow SSCs. However, after bone injuries, different populations of SSCs play distinct roles in bone repair, with periosteal cells primarily involved in non-stabilized fracture repair.

Interactions between FGF23 and vitamin D

Mohammed S. Razzaque

Summary: FGF23 controls the homeostasis of phosphate and vitamin D by suppressing the activation of vitamin D and enhancing its degradation process. There is a counter-regulation between FGF23 and vitamin D, with FGF23 inhibiting the synthesis of 1 alpha-hydroxylase (1 alpha(OH)ase) which produces vitamin D, while vitamin D induces the synthesis and release of FGF23. This feedback control is also observed in various human diseases.

Vitamin D Receptor Mediates a Myriad of Biological Actions Dependent on Its 1,25-Dihydroxyvitamin D Ligand: Distinct Regulatory Themes Revealed by Induction of Klotho and Fibroblast Growth Factor-23

Mark R. Haussler et al.

Summary: The hormonal vitamin D metabolite, 1,25-dihydroxyvitamin D [1,25(OH)(2)D], produced in the kidney, acts through the vitamin D receptor (VDR) in various end organs, regulating bone mineral homeostasis. VDR interacts with retinoid X receptor (RXR) to target vitamin D-responsive elements (VDREs) near regulated genes. Circulating levels of 1,25(OH)(2)D are controlled by PTH and FGF23, playing essential roles in calcium and phosphate balance.
