Lyophilization provides long-term stability for a lipid nanoparticle-formulated , nucleoside-modified mRNA vaccine

Summary: Research demonstrates that nucleoside-modified mRNA-LNP vaccines can be lyophilized and maintain their physicochemical properties after storage at room temperature for 12 weeks or at 4℃ for at least 24 weeks. Comparative studies also show that lyophilized mRNA-LNP vaccines maintain high expression and immunogenicity even after storage.

Hyaluronated and PEGylated Liposomes as a Potential Drug-Delivery Strategy to Specifically Target Liver Cancer and Inflammatory Cells

Summary: This study proposes the use of HA- and PEGylated-liposomes as potential treatments for hepatocellular carcinoma (HCC). The up-regulation of CD44 in HCC patients and a murine model suggests the involvement of this receptor in HCC progression. The experimental results show that HA-liposomes are more easily internalized by cells that over-express CD44, while PEG-modified liposomes are rapidly taken up by macrophages and induce M1 polarization in THP1 cells.

Differentiation of PC12 cell line into neuron by Valproic acid encapsulated in the stabilized core-shell liposome-chitosan Nano carriers

Summary: Valproic acid (VPA) was encapsulated in nano liposomes and stabilized by chitosan to improve its efficacy and reduce side effects. The results showed that chitosan enhanced the controlled release of VPA and had no toxicity effects on cells. The nano carriers also promoted the differentiation of PC12 cells into neurons, as validated by gene expression levels. Therefore, the Lip-VPA-Chit nano carrier has promising potential for treating neurodegenerative disorders.

Summary: mRNA and lipid nanoparticles have been studied as effective vaccine preparation systems against SARS-CoV-2 infection. However, the relationship between lipid nanoparticle composition and activity is not fully understood. In this study, researchers screened a panel of ionizable lipids and identified lipid nanoparticles with a branched-tail lipid structure as the most promising. Through buffer optimization and lyophilization, the mRNA vaccine was successfully stored and induced strong immune responses in mice and nonhuman primates, showing broad antibody activity against various virus strains.

Enhancing the preservation of liposomes: The role of cryoprotectants, lipid formulations and freezing approaches

Summary: This study investigated the formulation design for freeze-drying of two different liposomes and evaluated the role of various cryoprotectants in preserving their structural integrity. The findings showed that disaccharides and polyol successfully protected the liposomes, while the presence of surfactant during freezing process resulted in strong damage to the liposomes. Additionally, the choice of freezing rate depended on the composition of the lipid bilayer.
