Hypothalamic Kisspeptin Neurons: Integral Elements of the GnRH System

Vikash Prashar et al.

Summary: Highly complex interactions between different cells and hormonal signals are crucial for the reproductive competence of organisms. Kisspeptin neurons play a significant role in transmitting signals necessary for the onset of puberty and attainment of reproductive function, while also communicating with other neurons and nuclei. Dysfunction of kisspeptin and neurokinin B can lead to neuroendocrine disorders such as HH and infertility. Ca2+ signaling is essential for the generation of GnRH pulses, which are propagated through connexions between astrocytes and neurons. Impaired functioning of KNDy neurons may contribute to the development of PCOS.

Direct evidence that KNDy neurons maintain gonadotropin pulses and folliculogenesis as the GnRH pulse generator

Mayuko Nagae et al.

Summary: The study demonstrates that KNDy neurons serve as the GnRH pulse generator, and at least 20% of KNDy neurons are sufficient to maintain folliculogenesis. This provides a new therapeutic approach for treating infertile women.

Effect of electroacupuncture on the kisspeptin system in a pubertal rat model of polycystic ovary syndrome

Zhi Wang et al.

Summary: Electroacupuncture (EA) and sham acupuncture (SA) both showed improvements in symptoms of PCOS in pubertal rats by modulating the kisspeptin system in the hypothalamus. However, EA had a more comprehensive effect on ovarian function and PCOS improvement compared to SA.

Prenatal Androgen Exposure Alters KNDy Neurons and Their Afferent Network in a Model of Polycystic Ovarian Syndrome

Aleisha M. Moore et al.

Summary: PCOS, the most common endocrinopathy affecting women worldwide, is characterized by impaired negative feedback. This study explored the role of KNDy neurons in PCOS pathogenesis, revealing potential defects in negative feedback due to changes in gene expression within KNDy neurons and synaptic inputs from steroid hormone-responsive hypothalamic regions. Further research is needed to determine the specific contributions of these alterations to the neuroendocrine phenotype seen in PCOS.

Role of Kisspeptin on Hypothalamic-Pituitary-Gonadal Pathology and Its Effect on Reproduction

Jaskamal Padda et al.

Summary: Kisspeptin, a neuropeptide encoded by the KISS1 gene, plays a significant role in human reproduction by acting on the HPG axis. It is vital for reproductive system maturation and function, puberty onset, sexual behavior, ovum and sperm development, and pregnancy, and is implicated in disorders like PCOS and infertility. Kisspeptin levels can also help identify pregnancy-related complications and research is exploring its potential as a diagnostic marker and treatment for reproductive disorders.

Prenatal Androgen Exposure Alters KNDy Neurons and Their Afferent Network in a Model of Polycystic Ovarian Syndrome

Aleisha M. Moore et al.

Summary: Alterations in gene expression within KNDy neurons and reduced synaptic inputs from steroid hormone-responsive hypothalamic regions may contribute to defects in negative feedback in PCOS. The specific roles of these changes in contributing to the neuroendocrine phenotype seen in PCOS, as well as the involvement of specific sets of upstream KNDy afferents in the process, remain to be determined.
