Brothers in arms: proBDNF/BDNF and sAPPα/Aβ-signaling and their common interplay with ADAM10, TrkB, p75NTR, sortilin, and sorLA in the progression of Alzheimer's disease

Simone Eggert et al.

Summary: Brain-derived neurotrophic factor (BDNF) plays an important role in modulating various functions in the central nervous system (CNS), and its involvement in the progression of Alzheimer's disease (AD) is supported by evidence such as reduced levels of BDNF mRNA and protein in AD patients. The processing and localization of BDNF and its receptors TrkB and p75 are critical for neuronal cell survival and death. Similarly, the amyloid precursor protein (APP) and its cleavage fragments sAPP α and Aβ, which are key players in AD, have distinct roles in neuroprotection and neuronal death. The common features of APP and BDNF signaling suggest a causal relationship between them.

Decreased brain noradrenaline in minimal hepatic encephalopathy is associated with cognitive impairment in rats

Xuhong Yang et al.

Summary: This study investigated the levels of noradrenergic neurotransmitters in the hippocampus of a rat model with thioacetamide-induced chronic liver failure-related minimal hepatic encephalopathy (MHE), and their role in cognitive impairment. The results showed that MHE rats performed poorly in cognitive and behavioral tests, and exhibited iron staining and an imbalance of inflammatory cytokines in the hippocampus. Additionally, the level of noradrenaline was significantly lower in MHE rats compared to the control group.

SB332235, a CXCR2 antagonist, ameliorates thioacetamide-induced hepatic encephalopathy through modulation of the PI3K/AKT pathways in rats

Rania R. Abdelaziz et al.

Summary: This study evaluated the potential beneficial effects of the selective CXCR2 inhibitor SB332235 on hepatic encephalopathy (HE). The results demonstrated that SB332235 alleviated oxidative stress, reduced inflammation, protected brain function and liver injury, and activated the PI3K/AKT pathway, providing significant hepatic and neuroprotective effects.

Thymol ameliorated neurotoxicity and cognitive deterioration in a thioacetamide-induced hepatic encephalopathy rat model; involvement of the BDNF/CREB signaling pathway

Hanan A. Ogaly et al.

Summary: This study found that thymol has a neuroprotective effect against neurotoxicity and cognitive deterioration caused by hepatic encephalopathy. Thymol supplementation improved liver function, reduced serum ammonia levels, and improved locomotor and cognitive deficits. It modulated oxidative stress, neurotransmitters, and brain ATP levels, and ameliorated histopathological damage, astrocyte swelling, and brain edema. Thymol also downregulated NF-kB and upregulated GFAP expression, and promoted CREB and BDNF expression, as well as increased brain cAMP levels.

Naringenin attenuates endoplasmic reticulum stress, reduces apoptosis, and improves functional recovery in experimental traumatic brain injury

Chulei Deng et al.

Summary: Traumatic brain injury (TBI) is a major cause of disability and death globally, with accumulating evidence suggesting that endoplasmic reticulum (ER) stress plays a crucial role in its pathogenesis. Naringenin, a natural flavonoid, has been shown to ameliorate TBI-induced secondary brain injury by attenuating ER stress, oxidative stress, and apoptosis, indicating its potential for neuroprotective effects in TBI.
