Monte Carlo simulations of a lattice model of DNA origami revealed that some designs have a nucleation barrier, which disappears at lower temperatures. The height of the nucleation barrier depends on the coaxial stacking of adjacent staples, which can be modified with staple design. Creating or eliminating the nucleation barrier may have implications for optimizing assembly times, yields, and developing fast molecular sensors.

DNA Origami Voltage Sensors for Transmembrane Potentials with Single-Molecule Sensitivity

By using fluorescence resonance energy transfer (FRET) changes, voltage-induced displacement of anionic donor units was measured, showing a FRET change of about 5% for a membrane potential change of 100 mV. This approach demonstrates potential for non-genetically encoded membrane sensors.

RNA origami design tools enable cotranscriptional folding of kilobase-sized nanoscaffolds
