The Pseudomonas aeruginosa homeostasis enzyme AlgL clears the periplasmic space of accumulated alginate during polymer biosynthesis

Andreea A. Gheorghita et al.

Summary: This study investigates the structure and function of AlgL, an alginate lyase in Pseudomonas aeruginosa. Key active site residues, R249 and Y256, are identified. The results show that AlgL is necessary for clearing accumulated polymers and maintaining membrane integrity during alginate production. The AlgU/T sigma factor can compensate for an algL deletion. Overall, this study clarifies the role of AlgL in alginate biosynthesis.

Pseudomonas aeruginosa modulates alginate biosynthesis and type VI secretion system in two critically ill COVID-19 patients

Jiuxin Qu et al.

Summary: Our study suggests that SARS-CoV-2 infection can lead to genetic adaptation of bacterial pathogens, promoting chronic infections. The adapted bacteria exhibit characteristics such as excessive biofilm formation and reduced virulence, enabling them to evade immune clearance. This highlights the importance of understanding the lung microenvironment post-virus infections and superinfection mechanism in COVID-19 patients.

Quantifying Biomolecular Interactions Using Slow Mixing Mode (SLOMO) Nanoflow ESI-MS

Duong T. Bui et al.

Summary: Electrospray ionization mass spectrometry (ESI-MS) is a powerful label-free assay for detecting noncovalent biomolecular complexes and quantifying binding thermochemistry. However, the assumption that the ion signals of free and bound species in ESI-MS measurements can quantitatively reflect their relative concentrations is only valid when their response factors are similar.

UniProt: the universal protein knowledgebase in 2021

Alex Bateman et al.

Summary: The UniProt Knowledgebase aims to provide users with a comprehensive, high-quality set of protein sequences annotated with functional information. Updates over the past two years have increased the number of sequences to approximately 190 million, with new methods to assess proteome completeness and quality. UniProtKB has responded to the COVID-19 pandemic by expertly curating relevant entries and making them rapidly available through a dedicated portal.

Summary: The new cryo-EM structure of the ~1 MDa E. coli cellulose synthase macrocomplex reveals how cellulose biosynthesis and phosphoethanolamine modification are coupled to promote host-tissue adhesion. Through interactions between BcsA enzyme and BcsG subunit, the cellulose synthase machinery forms a complex molecular structure, providing detailed insights into cellulose synthesis.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.
