Highly activated CD4(+) cells emerged and dominated the CAR T cell population at later time points. These unexpected CAR T cell populations provide novel insights into anti-cancer response and long-term remission in leukaemia.

Chronologically modified androgen receptor in recurrent castration-resistant prostate cancer and its therapeutic targeting

Summary: Resistance to enzalutamide in castration-resistant prostate cancer (CRPC) is a common issue without effective therapeutic options. This study reveals that acetylation at Lys(609) in the DNA binding domain of androgen receptor (AR) allows AR translocation and confers enzalutamide insensitivity. The acetylation is mediated by ACK1 kinase-mediated AR Y267 phosphorylation, which leads to positive feedback loops at both transcriptional and posttranslational levels. Inhibiting ACK1 kinase with a small-molecule inhibitor compromises enzalutamide-resistant CRPC xenograft tumor growth.

EZH2 inhibits NK cell-mediated antitumor immunity by suppressing CXCL10 expression in an HDAC10-dependent manner

Summary: EZH2 has been shown to function as an oncogene in some cancers, mainly through regulating cell-intrinsic tumor regulatory pathways. However, the role of EZH2-mediated immune suppression, particularly regarding natural killer (NK) cells, in its oncogenic activity is not fully understood. In this study, it was revealed that EZH2 inhibition induced reexpression of chemokine CXCL10 in hepatic tumor cells, which in turn stimulated NK cell migration and reversed tumor growth. This suggests a crucial role for NK cells and CXCL10 in mediating the oncogenic function of EZH2.
