Peripheral serotonin: cultivating companionship with gut microbiota in intestinal homeostasis

Yun Han Kwon et al.

Summary: This review examines the interactions between serotonin and the gut microbiota, and discusses their importance for intestinal health. Recent studies have shown bidirectional interactions between the host serotonergic system and the gut microbiota, which may have implications for conditions such as inflammatory bowel disease.

IFN-γ+ cytotoxic CD4+ T lymphocytes are involved in the pathogenesis of colitis induced by IL-23 and the food colorant Red 40

Lili Chen et al.

Summary: Using single-cell transcriptomic analysis, researchers identified multiple subsets of CD4(+) T cells in the intestines of colitic mice, including a subset that produces interferon (IFN)-gamma. In vivo experiments showed that challenge with the food colorant Red 40 activated CD4(+) T cells and caused intestinal epithelial cell apoptosis, which was attenuated by depletion of CD4(+) cells and blocking IFN-gamma. Experiments conducted ex vivo demonstrated that intestinal CD4(+) T cells from colitic mice directly promoted apoptosis of intestinal epithelial cells and enteroids.

Salicylates Ameliorate Intestinal Inflammation by Activating Macrophage AMPK

Suhrid Banskota et al.

Summary: Genetic deletion of AMPK beta 1 in macrophages upregulates proinflammatory cytokine production, exacerbates dextran sodium sulfate-induced colitis severity in mice, increases nuclear translocation of nuclear factor-kappa B, and impairs autophagy both in vitro and in vivo. Additionally, commonly used anti-inflammatory drugs such as 5-aminosalicylic acid (mesalazine) and sodium salicylate ameliorate dextran sodium sulfate-induced colitis by activating macrophage AMPK targeting the beta 1 subunit.

Food colorants metabolized by commensal bacteria promote colitis in mice with dysregulated expression of interleukin-23

Zhengxiang He et al.

Summary: The study demonstrates that azo dyes Red 40 and Yellow 6 can induce IBD-like colitis in mice expressing IL-23, with the microbiota playing a role in metabolizing the dyes into harmful substances.

Disruption of autophagy by increased 5-HT alters gut microbiota and enhances susceptibility to experimental colitis and Crohn's disease

Sabah Haq et al.

Summary: This study revealed that gut 5-HT inhibited autophagy, leading to enhanced colitis susceptibility, while reduced 5-HT promoted autophagy and decreased colitis inflammation. Similar results were observed in control and patient blood samples, suggesting a potential therapeutic role of 5-HT in intestinal inflammatory disorders.

Low dietary fiber promotes enteric expansion of a Crohn's disease-associated pathobiont independent of obesity

Trevor C. Lau et al.

Summary: This study shows that lower dietary fiber is a key factor in promoting the expansion of gut pathobionts associated with Crohn's disease in mice, independent of obesity status. Reduction in dietary fiber was found to be sufficient to promote the intestinal expansion of pathogenic Escherichia coli, even without affecting body mass.

Early life exposures and the risk of inflammatory bowel disease: Systematic review and meta-analyses

Manasi Agrawal et al.

Summary: Early life exposures can impact the risk of immune-mediated diseases like inflammatory bowel disease, with factors such as prenatal exposure to antibiotics, tobacco smoke, and early otitis media potentially increasing the risk of IBD, while breastfeeding may have a protective effect. Other early life risk factors showed limited and heterogeneous associations with the disease.
