Polygenic Risk Scores Allow Risk Stratification for Keratinocyte Cancer in Organ-Transplant Recipients

Summary: Organ-transplant recipients have an elevated risk of keratinocyte cancers, particularly basal cell carcinoma and squamous cell carcinoma. Polygenic risk scores generated from nontransplantees can predict keratinocyte cancer risk in organ-transplant recipients, with the top 20% having significantly increased risks. Adding polygenic risk scores to traditional risk factors improves prediction of basal cell carcinoma by 3%.

MDA5 Governs the Innate Immune Response to SARS-CoV-2 in Lung Epithelial Cells

Summary: Recent studies show that cellular responses to SARS-CoV-2 infection may influence disease severity, with MDA5 and LGP2 primarily regulating IFN induction in lung epithelial cells. Further analysis revealed that viral intermediates specifically activate the IFN response through MDA5-mediated sensing, and transcription factors IRF3, IRF5, and NF-kappa B/p65 play key roles in regulating the IFN response during SARS-CoV-2 infection. These findings provide valuable insights into the molecular mechanisms of innate immune recognition and signaling response to SARS-CoV-2.

The incidence and clinical analysis of non-melanoma skin cancer

This study in Poland analyzed the incidence of primary BCCs and SCCs over a 20-year period, finding that BCC accounted for the majority of cases and SCC patients were typically older. Polygenic risk scores can improve the prediction of BCC and SCC risks, especially in SOTRs with high genetic risk. The polygenic risk scores also add predictive accuracy for tumor multiplicity in this population.
