Tie2 Activation via VE-PTP Inhibition With Razuprotafib as an Adjunct to Latanoprost in PatientsWith Open Angle Glaucoma or Ocular Hypertension

Mitchell Brigell et al.

Summary: The study investigated the ocular hypotensive efficacy and safety of razuprotafib, a Tie2 activator, as an adjunct to latanoprost in patients with open-angle glaucoma or ocular hypertension. It was found that using razuprotafib twice daily with latanoprost significantly reduced intraocular pressure (IOP) compared to latanoprost monotherapy. However, razuprotafib once daily did not provide additional IOP lowering. The study also showed that razuprotafib therapy was well tolerated but could cause conjunctival hyperemia.

Analysis of 200 000 exome-sequenced UK Biobank subjects illustrates the contribution of rare genetic variants to hyperlipidaemia

David Curtis

Summary: Rare variants in genes, particularly on the X chromosome, have been found to have major effects on lipid levels. The study demonstrated the importance of including principal components and sex as covariates in genetic analysis, and identified significant associations with genes like LDLR and PCSK9. Variants classified as deleterious by SIFT were found to have a twofold effect and are present in approximately 1.5% of the population, highlighting their impact on hyperlipidaemia risk.

Cell atlas of the human ocular anterior segment: Tissue-specific and shared cell types

Tave van Zyl et al.

Summary: In this study, we generated an anterior segment cell atlas of the human eye using high-throughput single-nucleus RNA sequencing (snRNAseq) and identified over 60 cell types. The atlas revealed the cell type characteristics of different tissues in the anterior segment and the sharing of certain cell types across tissues. Additionally, the atlas was used to investigate cell type-specific expression patterns of genes associated with ocular diseases.

Exome sequencing and analysis of 454,787 UK Biobank participants

Joshua D. Backman et al.

Summary: In this study, whole-exome sequencing was used to identify gene-trait associations in 454,787 individuals, revealing 564 distinct genes with significant trait associations. Rare variant associations were enriched in loci from genome-wide association studies (GWAS) but most were independent of common variant signals.

Summary: The authors conducted a meta-analysis of genome-wide association studies in 34,179 cases of POAG, identifying 44 previously unreported risk loci and mapping effects across multiple ethnicities. Integration of multiple lines of genetic evidence supports the functional relevance of the identified POAG risk loci.

Rare variant contribution to human disease in 281,104 UK Biobank exomes

Quanli Wang et al.

Summary: The study reveals a significant contribution of rare variants to common disease, with a large number of gene-phenotype associations detected through gene-based collapsing analysis that cannot be identified in single-variant association tests. Rare variants are also significantly enriched for loss-of-function-mediated traits and approved drug targets.
