Inosine and its methyl derivatives: Occurrence, biogenesis, and function in RNA

Summary: Inosine is a common post-transcriptional modification in RNA, which can contribute to non-Watson-Crick interactions. The generation of inosine through hydrolytic deamination of adenosine is widespread, and its methyl derivatives have been reported in RNA modification databases. These modifications can lead to changes in RNA's structure, folding, dynamics, stability, and functions. The conservation of modifying enzymes across organisms indicates the importance of these modifications. The mechanisms underlying the catalytic reaction of adenosine deaminases are not clear yet. Further studies on the occurrence, biogenesis, and functions of inosine and its methyl derivatives in RNA, as well as the structural and thermodynamic aspects of these modifications, are crucial for understanding their role in different diseases.
