Opto-vTrap, an optogenetic trap for reversible inhibition of vesicular release, synaptic transmission, and behavior

Joungha Won et al.

Summary: Spatiotemporal control of brain activity can be achieved using Opto-vTrap, which is a light-inducible and reversible system that temporarily inhibits the release of neurotransmitters. Opto-vTrap significantly reduces synaptic and gliotransmission, leading to inhibition of hippocampus-dependent memory retrieval.

Optogenetics at the presynapse

Benjamin R. Rost et al.

Summary: This review provides a comprehensive overview of the presynaptic applications of optogenetic tools, discussing the challenges, limitations, and future directions. Optogenetic actuators enable precise control of presynaptic functions, such as light-mediated neurotransmitter release, presynaptic inhibition, induction of synaptic plasticity, and manipulation of individual components of the presynaptic machinery.

Dynamin is primed at endocytic sites for ultrafast endocytosis

Yuuta Imoto et al.

Summary: This study demonstrates that Dynamin 1 is pre-recruited to endocytic sites for ultrafast endocytosis. Syndapin 1 acts as an adaptor by binding the plasma membrane and storing Dynamin 1xA at endocytic sites, bypassing the recruitment step and accelerating endocytosis. These findings reveal the mechanism of endocytosis and provide new insights into cellular processes.

Optogenetic Stimulation of Gi Signaling Enables Instantaneous Modulation of Cardiomyocyte Pacemaking

Milan Cokic et al.

Summary: G-protein signaling pathways play a crucial role in regulating cardiac function, and optogenetic techniques utilizing opsin proteins allow precise activation in cardiomyocytes. The application of long wavelength-sensitive cone opsin (LWO) in this study demonstrated effective and specific activation of the G(i) signaling cascade in cardiomyocytes with high temporal precision.

Synapsin Is Required for Dense Core Vesicle Capture and cAMP-Dependent Neuropeptide Release

Szi-Chieh Yu et al.

Summary: The release of neuropeptides is crucial for neuromodulation, and our study demonstrates that synapsin SNN-1 is necessary for cAMP-dependent neuropeptide release in Caenorhabditis elegans. Through capturing dense core vesicles (DCVs) at synapses, synapsin plays a key role in making them available for release. Mutations in synapsin result in altered neuropeptide release and behavior in the nematode, indicating the importance of synapsin in this process.

Efficient optogenetic silencing of neurotransmitter release with a mosquito rhodopsin

Mathias Mahn et al.

Summary: A mosquito homolog of vertebrate encephalopsin was found to effectively suppress synaptic transmission through a specific signaling pathway, triggering lasting synaptic output inhibition that recovers spontaneously in vitro and in vivo. This discovery opens up new avenues for functional exploration of long-range neuronal circuits in vivo.

A photoswitchable GPCR-based opsin for presynaptic inhibition

Bryan A. Copits et al.

Summary: This study introduces a GPCR-based opsin called parapinopsin (PPO), which can rapidly and reversibly inhibit the release of glutamate, GABA, and dopamine at presynaptic terminals by coupling with G(i/o) signaling cascades. PPO alters reward behaviors in a time-locked and reversible manner in vivo, filling a significant gap in the neuroscience toolkit for rapid and reversible synaptic inhibition.
