Photochemical Probe Identification of a Small-Molecule Inhibitor Binding Site in Hedgehog Acyltransferase (HHAT)**

Summary: This study reports the rational development of a photochemical probe to investigate a novel small-molecule inhibitor binding site in the human MBOAT Hedgehog acyltransferase (HHAT), leading to the identification of the most potent HHAT inhibitor IMP-1575 to date. Through structure-activity relationship investigation and methods such as photocrosslinking, the study revealed the inhibition mechanism and small molecule binding site of HHAT, providing a strategy for mapping small molecule interaction sites in MBOATs.
