Lipid nanoparticle delivery of unmodified mRNAs encoding multiple monoclonal antibodies targeting poxviruses in rabbits

Eric M. Mucker et al.

Summary: This study explored the feasibility of using unmodified mRNA encoding three previously described monoclonal antibodies as countermeasures against smallpox in rabbits. The researchers confirmed the translation, secretion, and biological activity of the mRNA constructs in vitro and found that injecting a combination of the three mRNA constructs produced similar serum levels as individual injections. This is the first evidence of launching multiple antibodies using mRNA constructs in a large nonrodent species.

A human recombinant analogue to plasma-derived vaccinia immunoglobulin prophylactically and therapeutically protects against lethal orthopoxvirus challenge

Scott Parker et al.

Summary: Orthopoxviruses such as variola and monkeypox viruses pose a continuing threat to the human population, with the need for antiviral drugs in case of outbreaks or deliberate dissemination. While the U.S. stockpiles two antivirals for smallpox treatment, other options like rVIG show efficacy in treating various orthopoxviruses.

New methylene blue derivatives suggest novel anti-orthopoxviral strategies

Lalita Priyamvada et al.

Summary: Decades after smallpox eradication and cessation of routine vaccination, over half of the world's population lacks immunity to variola virus and other orthopoxviruses, leading to an increase in human OPXV infections. Methylene blue derivatives, including PAV-866, exhibit potent inhibitory activity against vaccinia virus, showcasing a novel mode of inhibition and potential for combating OPXVs.
