VIGS Goes Viral: How VIGS Transforms Our Understanding of Plant Science

Clemens Roessner et al.

Summary: Virus-induced gene silencing (VIGS) is an indispensable method for gene function analysis, especially in species not amenable to genetic transformation. It controls viral infections by utilizing the plant's gene silencing mechanism and is widely used for gene downregulation. Recently, VIGS technology has been expanded to achieve gene overexpression, genome editing, and host-induced gene silencing.

Petabase-scale sequence alignment catalyses viral discovery

Robert C. Edgar et al.

Summary: Public databases contain a vast amount of nucleic acid sequences, but efficient methods for systematic exploration of this library have been lacking. In this study, we developed a cloud computing infrastructure, Serratus, to enable ultra-high-throughput sequence alignment at the petabase scale. By searching millions of diverse samples, we identified over 10(5) novel RNA viruses and characterized their environmental reservoirs. To facilitate viral discovery, we established a comprehensive database of these data and tools. Expanding the known sequence diversity of viruses has important implications for understanding emerging pathogens and improving pathogen surveillance.

Genome-edited powdery mildew resistance in wheat without growth penalties

Shengnan Li et al.

Summary: Disruption of susceptibility genes in crops can confer disease resistance, but often leads to undesired effects. This study identifies a mutant wheat variety with a targeted deletion in a specific gene that retains crop growth and yields while conferring robust resistance to powdery mildew.

Biomimetic Virus-Like Particles as Severe Acute Respiratory Syndrome Coronavirus 2 Diagnostic Tools

Soo Khim Chan et al.

Summary: COVID-19 is a highly transmissible disease affecting majority of countries globally. The use of nucleic acid tests, especially RT-PCR, has become essential for early detection of infection. Generating biomimetic virus-like particles as positive controls can overcome the need for cold-chain distribution, providing reliable, scalable, and stable controls for clinical settings.

Agricultural land use disrupts biodiversity mediation of virus infections in wild plant populations

Hanna Susi et al.

Summary: The study found that plant species richness and diversity were negatively correlated with virus infection prevalence. Virus species richness decreased with increasing plant diversity and richness in natural populations, while in agricultural edge populations, virus species richness was moderately higher and not associated with plant richness.

Plant virus evolution under strong drought conditions results in a transition from parasitism to mutualism

Ruben Gonzalez et al.

Summary: Environmental conditions, such as drought stress, play a significant role in the evolution of pathogens like plant viruses. Viruses evolved under drought conditions can enhance the drought tolerance of infected plants, with differences in tolerance linked to changes in host gene expression and phytohormone balance. This study demonstrates that viruses can help host survival in abiotic stress situations, and the extent of this benefit is a selectable trait.

Current Developments and Challenges in Plant Viral Diagnostics: A Systematic Review

Gajanan T. Mehetre et al.

Summary: Plant viral diseases pose a significant threat to sustainable agriculture, and timely and accurate identification of causative agents is crucial for effective treatment. Current diagnostic methods primarily involve serological and nucleic acid techniques, but there is a growing interest in developing real-time diagnostic tools to improve in-field diagnosis capabilities. Next-generation sequencing (NGS) shows great potential for detecting multiple viruses simultaneously, but further research is needed to optimize these techniques for plant virology.

Application of Oxford Nanopore Technology to Plant Virus Detection

Lia W. Liefting et al.

Summary: The adoption of Oxford Nanopore Technologies (ONT) sequencing in plant virology has been slow, but its promise of real-time generation of long nucleotide sequences makes it an attractive method for both low- and high-scale virus diagnostics. The portability of the MinION and Flongle platforms, combined with lower costs and improved read accuracy, highlight ONT's feasibility as a valuable component to diagnostician's toolkit.

Transient reprogramming of crop plants for agronomic performance

Stefano Torti et al.

Summary: The proposed technology targets regulatory circuits within plants transiently, allowing for customized alterations of agronomic traits. By utilizing RNA viral transfection methods, this approach is fast, versatile, and does not involve genetic modification of plant genomes. It can be used in multiple plant species for rapid fine-tuning of traits without altering the genome.
