EBV infection-induced GPX4 promotes chemoresistance and tumor progression in nasopharyngeal carcinoma

Li Yuan et al.

Summary: This study found that Epstein-Barr virus (EBV) infection reduces the sensitivity of nasopharyngeal carcinoma (NPC) cells to ferroptosis by activating the p62-Keap1-NRF2 signaling pathway and upregulating SLC7A11 and GPX4 expression. Inhibition of GPX4 enhances the chemosensitivity of EBV-infected NPC cells. High GPX4 expression is associated with poor clinical outcomes in NPC and other cancer types.

Epstein-Barr virus latency programs dynamically sensitize B cells to ferroptosis

Eric M. Burton et al.

Summary: Epstein-Barr virus (EBV) infection plays a role in multiple cancers, and this study examines how EBV infection affects sensitivity to ferroptosis, a form of cell death driven by lipid peroxidation and membrane damage. The study finds that EBV-infected B cells are sensitive to blockage of glutathione biosynthesis and have elevated levels of lipid reactive oxygen species (ROS) that require SLC7A11 and GPX4 activity to prevent ferroptosis. Different stimulation methods affect the sensitivity of B cells to ferroptosis, and EBV-transformed B cells become progressively resistant to ferroptosis induction.

Swine influenza virus triggers ferroptosis in A549 cells to enhance virus replication

Jinghua Cheng et al.

Summary: This study reveals a new mode of cell death induced by IAV infection, known as ferroptosis. The results show that SIV infection disrupted iron homeostasis and transport, and inhibited the Xc(-)/GPX4 axis, leading to depletion of glutathione and accumulation of lipid peroxidation products. Additionally, the study found that using a ferroptosis inhibitor partially rescued cell death caused by iron-dependent lipid peroxidation.

NCOA4-mediated ferritinophagy promotes ferroptosis induced by erastin, but not by RSL3 in HeLa cells

Magdalena Gryzik et al.

Summary: This study found that NCOA4 deficiency increases ferritin levels and makes cells more resistant to erastin but more sensitive to RSL3. Erastin promotes ferritinophagy, increasing free iron, lipid peroxidation, and sensitivity to ferroptosis, while RSL3 mechanism is independent of ferritin degradation process. The release of ferritin-iron in the execution of ferroptosis seems to depend on the inducing compound, its target, and downstream pathway of cell death activation.
