Interferon-Gamma Primed Human Clonal Mesenchymal Stromal Cell Sheets Exhibit Enhanced Immunosuppressive Function

Celia M. Dunn et al.

Summary: Mesenchymal stromal cells (MSCs) have diverse immunomodulatory functions and hold promise as a treatment for immune-related diseases. In this study, IFN-γ priming and thermoresponsive cultureware were combined to produce transplantable scaffold-free immunosuppressive cell sheets. The use of human clinical-grade single-cell-derived clonal bone marrow-derived MSCs further enhances the clinical relevance of this approach. The results show that MSC sheets produced with IFN-γ priming upregulate expression of immunosuppressive factors and exhibit increased ability to inhibit T-cell proliferation.

Allogeneic mesenchymal stem cell sheet therapy: A new frontier in drug delivery systems

Kyungsook Kim et al.

Summary: The evolution of drug discovery in the early 20th century saw a shift from focusing on symptom control to pursuing radical cures, with cell therapy emerging as a promising new treatment targeting specific tissues and dynamic signaling environments. Cell delivery methods play a crucial role in realizing the therapeutic potential of cell therapy, especially in the context of mesenchymal stem cell (MSC) engraftment and bioactive function. Cell sheet technology is presented as a breakthrough in cell therapy, showcasing therapeutic success in tissue regeneration and discussing its anticipated clinical impacts.

Renal subcapsular delivery of PGE2 promotes kidney repair by activating endogenous Sox9+ stem cells

Shang Chen et al.

Summary: The study found that by covalently cross-linking PGE(2) to a collagen matrix, it can achieve long-term presence in the kidney, significantly improve kidney function, and activate endogenous renal progenitor cells. The research highlights the potential of renal subcapsular-based drug delivery and expands the applications of PGE(2)-releasing matrices in regenerative therapy.

3D cell sheet structure augments mesenchymal stem cell cytokine production

Sophia Bou-Ghannam et al.

Summary: This study demonstrated that mesenchymal stem cells (MSCs) can enhance their therapeutic potency through cultivation in a scaffold-free cell sheet technology to form 3D tissue-like structures. The transition from 2D to 3D culture promoted changes in cell morphology, cytoskeleton organization, and gene expression of pro-tissue regenerative cytokines, ultimately increasing MSC secretion of key growth factors.
