Low-Dose Radiotherapy Leads to a Systemic Anti-Inflammatory Shift in the Pre-Clinical K/BxN Serum Transfer Model and Reduces Osteoarthritic Pain in Patients

Thomas Weissmann et al.

Summary: Osteoarthritis is a common degenerative joint disease, particularly affecting the ankle and foot. Low-dose radiotherapy has been shown to be highly effective in treating osteoarthritis, with analgesic effects mediated by an interplay of cellular and immune factors, especially in younger patients.

ROS- and Radiation Source-Dependent Modulation of Leukocyte Adhesion to Primary Microvascular Endothelial Cells

Summary: Low-dose irradiation has anti-inflammatory effects on endothelial cells, and the dose-effect relationship is non-linear. Laminar conditions contribute to increased expression of anti-oxidative factors and reduced intracellular ROS content, resulting in reduced leukocyte adhesion.

Summary: X-ray irradiation inhibits the differentiation of precursor osteoclasts into mature osteoclasts and reduces their bone resorbing activity. The decreased NFATc1 signaling may be one underlying mechanism.

Obesity, Inflammation, and Immune System in Osteoarthritis

Udhaya Nedunchezhiyan et al.

Summary: Obesity is the most important risk factor for the development and progression of osteoarthritis, and it was previously believed to be caused by excessive weight leading to joint overloading and cartilage destruction. However, recent studies have identified other factors such as adipose deposition, insulin resistance, and the improper coordination of innate and adaptive immune responses that contribute to obesity-associated osteoarthritis. Multiple inflammatory cells are recruited into the synovial joint and play an important role in pathological changes. Macrophage polarization and mediators produced by macrophages have been extensively studied and linked to the inflammatory and destructive responses in the synovium and cartilage. The role of other innate immune cells and adaptive immune cells in the pathogenesis of osteoarthritis is still under exploration.

Summary: Ionizing radiation interacts with the immune system in complex ways, potentially altering immune fitness and accelerating immune senescence at low doses. However, low dose radiation therapy has beneficial effects in certain diseases, providing relief from pain and inflammation.

Summary: Osteoarthritis is a prevalent joint disease that can impact various joints, with risk factors including age, gender, obesity, genetics, and major joint injury. Treatment typically involves exercise, weight management, education, and medication therapy. Advanced cases may require surgical intervention for pain relief and improved function.

Summary: LDRT shows promise in treating chronic inflammatory and degenerative diseases, improving pain levels and inducing systemic immune modulations; LDRT leads to a slight reduction in certain immune cell populations and an increase in B cells; Monitoring immune biomarkers may help predict therapeutic success and symptom improvement in LDRT.

Fibroblast-like synoviocytes in rheumatoid arthritis: Surface markers and phenotypes

Zewen Wu et al.

Summary: Fibroblast-like synoviocytes (FLS) play a crucial role in the pathophysiology of rheumatoid arthritis (RA) and have become the main focus of research. Advances in technology, such as high-throughput sequencing, have made it possible to identify different FLS subsets, allowing for a detailed study of RA pathogenesis.

Low-dose radiation therapy for osteoarthritis and enthesopathies: a review of current data

Seyed Alireza Javadinia et al.

Summary: Research has shown the clinical benefits of low-dose radiotherapy in managing osteoarthritis, such as pain relief, improved mobility and function, with minimal side effects. Mechanistically, it involves the activation of the nuclear factor erythroid 2-related transcription factor (Nrf2) mediated antioxidant response, leading to positive subcellular effects. Further studies on the short- and long-term effects of low-dose radiotherapy on osteoarthritis and other inflammatory disorders are recommended.

Summary: The study showed that low-dose radiotherapy is effective in reducing pain for elderly patients with osteoarthritis, with no difference in pain response rates as age increases. Additionally, there was no significant variation in pain response after re-irradiation across different age groups.

Andres Coca-Pelaz et al.

Summary: This study found that the frequency of radiation-induced sarcomas of the head and neck (RISHN) is relatively low, with the most common primary tumor location treated by RT being the nasopharynx and the most common RISHN location being the sinonasal region. Surgery is the main treatment modality, but the prognosis remains poor.
