Circular Dichroism Spectra of α-Chymotrypsin-SDS Solutions Depend on the Procedure of Their Preparation

Summary: In this study, the interaction between chymotrypsin and SDS at different concentrations was investigated by recording the CD spectra of the solutions. It was found that supplementing the chymotrypsin solution with SDS to achieve the same final concentration led to different CD spectra compared to directly adding SDS to the chymotrypsin solution. These differences disappeared when the initial concentration of SDS in the protein solution was higher, indicating the irreversibility of the SDS addition process and the influence of the initial SDS amount on subsequent addition. Quantitative analysis of the CD spectra revealed an increase in the population of helical forms in chymotrypsin under the influence of SDS.

Stability comparison of four lipases and catalytic mechanism during the synthesis of 1,3-di-oleic-2-medium chain triacylglycerols in a trace water-in-oil system: Experimental analyses and computational simulations
