Silver nanoclusters show advantages in macrophage tracing in vivo and modulation of anti-tumor immuno-microenvironment

Summary: Macrophage-based nanomedicine is a promising strategy for cancer therapy, but there are obstacles and challenges to its translational applications. This study reports a novel type of silver nanoclusters (AgNCs) that have excellent fluorescent intensity and long-lasting cell labeling stability after being taken up by macrophages. The AgNCs escape lysosomal degradation within macrophages and stimulate M1-like polarization of macrophages through the toll-like receptor 4 signaling pathway. The engineered macrophages loaded with AgNCs can effectively target lung metastasis breast cancer and regulate the tumor immune microenvironment, leading to tumor growth inhibition.
