Megabodies expand the nanobody toolkit for protein structure determination by single-particle cryo-EM

Summary: Nanobodies are versatile tools for structural biology that can help overcome obstacles in single-particle cryo-electron microscopy reconstructions. Engineered megabodies, which have increased molecular weight while retaining specificity and affinity, are applicable to different scaffold proteins and recognition domains, and are useful for obtaining three-dimensional reconstructions for membrane proteins with severe preferential orientation.

Preparing Better Samples for Cryo-Electron Microscopy: Biochemical Challenges Do Not End with Isolation and Purification

Summary: Transmission electron microscopy poses extreme risk of damaging biological macromolecules due to interactions with the air-water interface. Current research is focused on finding methods to substantially reduce this hazard, primarily through immobilizing particles and reducing interaction time. There is also a growing focus on gaining more effective control of sample thickness before vitrification.

A new method utilizing small-molecule ligands for affinity capture of human target proteins directly onto grid surfaces has been demonstrated. This approach enables structure elucidation of multiple configurational states of the target proteins, potentially aiding structure-based drug design.
