Construction of emissive ruthenium(II) metallacycle over 1000 nm wavelength for in vivo biomedical applications

Fabrication of a phototheranostic nanoplatform for single laser-triggered NIR-II fluorescence imaging-guided photothermal/chemo/antiangiogenic combination therapy

In this study, a new small molecule dye and a redox-responsive prodrug were designed and synthesized to fabricate a multifunctional phototheranostic nanoplatform. The nanoplatform allows high-resolution imaging and efficient photothermal therapy in the second near-infrared window, and the prodrug can be cleaved in the tumor microenvironment to release the chemotherapeutic drug and angiogenesis inhibitor. Laser-induced hyperthermia was utilized to control the drug release and enhance the therapeutic effect. The developed nanoplatform showed improved antitumor efficacy in vitro and in vivo.

Organic single molecule based nano-platform for NIR-II imaging and chemo-photothermal synergistic treatment of tumor

Summary: In this study, five novel NIR-II dyes were successfully developed with intense absorption ranging from 890 to 1206 nm, strong singlet oxygen generation capability, and fluorescence imaging ability triggered by a single NIR-II light. Among them, BH 1024 showed the best performance in singlet oxygen generation, moderate photothermal heating, and considerable fluorescence under 1064 nm laser irradiation. Encapsulating BH 1024 into folate-functionalized polymer demonstrated a synergetic PDT/PTT effect in vitro and in vivo, eventually achieving solid tumors elimination under NIR-II fluorescence guide. This is the first attempt to explore and design small molecular dyes for NIR-II PDT or NIR-II PDT/PTT.
