Whole-genome sequencing of East Asian lung cancers reveals new germline pathogenic variants

Semanti Mukherjee et al.

Summary: In this study, Wang et al. used whole-genome sequencing to comprehensively analyze common and rare variants associated with lung cancer risk in individuals with East Asian ancestry. Their findings suggest that rare promoter variants in the BRCA2 gene are associated with an increased risk of lung cancer.

Metabolic adaptation of lymphocytes in immunity and disease

Nicole M. Chapman et al.

Summary: Adaptive immune responses mediated by T cells and B cells are crucial for protective immunity against pathogens and tumors. Understanding how metabolic plasticity and adaptation dictate functional specialization of immune cells is fundamental to our understanding and therapeutic modulation of the immune system. Extensive progress has been made in characterizing the effects of metabolic networks on immune cell fate and function in different immunological contexts.

Mitochondrial electron transport chain is necessary for NLRP3 inflammasome activation

Leah K. Billingham et al.

Summary: The study found that inhibitors of different complexes in the mitochondrial electron transport chain (ETC) can prevent the activation of the NLRP3 inflammasome, and the expression of specific enzymes from yeast or tunicates can rescue the activation of NLRP3 inflammasome in the absence of functional mitochondrial complex I or III. Metabolomics revealed that ATP generation through the phosphocreatine-dependent mechanism sustains the activation of NLRP3 inflammasome.

Lactate supports a metabolic-epigenetic link in macrophage polarization

Jordan T. Noe et al.

Summary: Lactate accumulation plays a key role in the activation of M2 macrophages, promoting histone acetylation, M2 gene transcription, and functional immune suppression. Metabolism of M2 macrophages is dependent on mitochondrial function and the glucose/lactate metabolic pathway.

Energetic dysfunction in sepsis: a narrative review

Sebastien Preau et al.

Summary: Organ dysfunction in sepsis is associated with impaired metabolism and mitochondrial dysfunction, which could lead to new diagnostic and therapeutic approaches. These findings may help identify distinct subgroups of sepsis and develop personalized treatment strategies, while also serving as bioenergetic targets for metabolism- and mitochondria-targeted treatments in the future.
