Fullerene nanoparticles for the treatment of ulcerative colitis

Xiaodan Liao et al.

Summary: Ulcerative colitis (UC) is a chronic inflammatory bowel disease without an effective cure, but a novel therapeutic strategy using C-60 fullerene suspension (C60FS) has shown promising results in an animal model. C60FS can repair colon barrier dysfunction, promote ulcer healing, reduce inflammation, and inhibit the expression of certain receptors and inflammatory markers in colorectal tissues. This study provides a reliable strategy based on fullerene for treating UC and offers a novel approach for UC therapy.

Micro-Coevolution of Genetics Rather Than Diet With Enterotype in Pigs

Ning Ma et al.

Summary: This study introduces the enterotype model to decipher the colonization and transition of gut microbiota in Chinese pig breeds. Based on host genetics, three representative enterotype clusters were identified, and Chinese native breeds were found in two of them. The study also isolated Lactobacillus reuteri-E3 as a representative strain, which exhibited strong stress resistance and potential intervention in weaned piglet diseases.

Dietary nutrition regulates intestinal stem cell homeostasis

Ning Ma et al.

Summary: This article reviews the critical role of intestinal stem cells (ISCs) in governing the proliferation and differentiation of the intestinal epithelium, as well as the regulation of ISCs by dietary factors. Through pathways such as mTORC1, PPAR/CPT1 alpha, PPAR gamma/beta-catenin, and Wnt/GSK-3 beta, factors such as caloric restriction, fasting, fat, glucose, and trace elements in the diet affect the function and self-renewal of ISCs, and may serve as potential targets for cancer prevention. However, a lack of precise definitions of nutrition intervention hinders our understanding of the biological response of ISCs to nutrients.

TRIM34 attenuates colon inflammation and tumorigenesis by sustaining barrier integrity

Qiaoshi Lian et al.

Summary: This study identifies a novel function of TRIM34 in goblet cells in controlling the generation of the colonic inner mucus layer, suggesting that TRIM34 deficiency leads to defects in the inner mucus layer and increased susceptibility to colitis and colorectal cancer. Mechanistically, TRIM34 controls TLR signaling-induced ROS synthesis and promotes the exocytosis of Muc2, demonstrating its crucial role in preventing excessive colon inflammation and tumorigenesis. Clinical analysis further reveals a correlation between TRIM34 levels and the prognosis of ulcerative colitis and rectal adenocarcinoma.

Construction of a sustainable 3-hydroxybutyrate-producing probiotic Escherichia coli for treatment of colitis

Xu Yan et al.

Summary: Colitis is a difficult-to-treat disease of the colon. Probiotic bacteria, specifically engineered Escherichia coli Nissle 1917, have been used to produce the ketone body (R)-3-hydroxybutyrate (3HB) in the gut of mice with colitis, leading to improved gut health and potential treatment of the disease.

Foxo1 controls gut homeostasis and commensalism by regulating mucus secretion

Zuojia Chen et al.

Summary: Foxo1 protein regulates mucus secretion by goblet cells to maintain intestinal homeostasis. Loss of Foxo1 leads to defects in autophagy and mucus secretion by goblet cells, impacting gut microbiota and causing increased susceptibility to intestinal inflammation.

Design, optimization, and nanotechnology of antimicrobial peptides: From exploration to applications

Peng Tan et al.

Summary: The abuse of antibiotics causing antibiotic resistance is a serious threat to human health, and antimicrobial peptides have become potential antibiotic alternatives due to their unique mechanisms. Despite the problems with natural AMPs, advancements in molecular design, optimization strategies, and nanotechnology have shown potential to improve their properties. Progress in the design and optimization strategies of AMPs lay a theoretical foundation for the development of peptide-based antimicrobial nanomaterials, providing more effective antibiotic alternatives.
