The neurobiology of treatment-resistant depression: A systematic review of neuroimaging studies

Nora Runia et al.

Summary: Our systematic review of neuroimaging studies reveals neurobiological differences between patients with Treatment-resistant depression (TRD) and non-TRD, including reduced functional connectivity within the default mode network (DMN), reduced connectivity between DMN and other brain areas, and hyperactivity of DMN regions. Aberrant activity and connectivity in the occipital lobe may also play a role in TRD. However, limitations in existing studies highlight the need for prospective longitudinal neuroimaging designs to better understand the effects of disease progression in TRD.

Altered Brain Function in Treatment-Resistant and Non-treatment-resistant Depression Patients: A Resting-State Functional Magnetic Resonance Imaging Study

Jifei Sun et al.

Summary: In this study, the differences in local brain functional activity and its characteristics were observed in patients with treatment-resistant depression (TRD) and non-treatment-resistant depression (nTRD) using amplitude of low-frequency fluctuation (ALFF) and regional homogeneity (ReHo). The study found that abnormal neurological functional activity was present in some brain regions, and these abnormalities were correlated with clinical symptoms.

Distinct neural mechanisms of emotional processing in prolonged grief disorder

Richard A. Bryant et al.

Summary: This study investigated distinctive neural processes underpinning emotion processing in participants with Prolonged Grief Disorder (PGD) compared to Posttraumatic Stress Disorder (PTSD) and Major Depressive Disorder (MDD). The results showed that PGD had unique neural activation patterns, particularly during processing of happy and sad faces, compared to PTSD and MDD groups. These findings provide initial evidence of distinct neural profiles of PGD and highlight the activation of neural regions implicated in reward networks.

Sub-regional anterior cingulate cortex functional connectivity revealed default network subsystem dysfunction in patients with major depressive disorder

Xiaolong Peng et al.

Summary: The study identified significant changes in functional connectivity (FC) of different anterior cingulate cortex (ACC) sub-regions in major depressive disorder (MDD) patients, associated with distinct symptoms of depression. The findings provide new insights into the role of ACC sub-regions and the default network (DN) in the pathophysiology of MDD.

Cross-network interaction for diagnosis of major depressive disorder based on resting state functional connectivity

Summary: This study suggests that cross-network interaction can serve as an effective biomarker for the clinical diagnosis of MDD, potentially revealing the underlying pathological mechanism for major depression. It also confirms the reliable application of MVPA in discriminating MDD patients from healthy controls.

Decreased dynamism of overlapping brain sub-networks in Major Depressive Disorder

Nan Chen et al.

Summary: The study found that patients with Major Depressive Disorder (MDD) show significantly decreased dynamism in certain sub-networks related to Visual, Default Mode, Cognitive Control, Bilateral Limbic, and Auditory networks compared to healthy controls. The overlapping brain regions in MDD patients may be affected by alterations in hubs. Further statistical analysis revealed decreased dynamism in five sub-networks in MDD patients, providing insights into the mechanisms of MDD.

Abnormal functional connectivity based on nodes of the default mode network in first-episode drug-naive early-onset schizophrenia

Summary: Schizophrenia is considered as a connectivity disorder, with research on the functional connectivity of the default mode network (DMN) lacking in early-onset schizophrenia. The study found abnormal connectivity within the DMN and between DMN and visual networks in early-onset schizophrenia patients, suggesting the involvement of DMN in the pathogenesis of EOS.

Summary: Somatic depression is more severe than pure depression. Reduction in ReHo and ALFF in the precentral gyrus, postcentral gyrus, and paracentral gyrus may play a significant role in the pathophysiology of somatic symptoms in MDD.

Altered resting-state functional connectome in major depressive disorder: a mega-analysis from the PsyMRI consortium

Nooshin Javaheripour et al.

Summary: This study investigated functional alterations in MDD patients, finding significant hypoconnectivity within SMN and SN networks, as well as between various networks. Differences in network organization were observed, with MDD patients showing lower network segregation in SMN. Although medicated patients had lower FC within certain networks, network organization differences were not significant between medicated and unmedicated groups, suggesting that network organization of cortical networks may be a more stable neuroimaging marker for MDD.

Resting state functional connectivity subtypes predict discrete patterns of cognitive-affective functioning across levels of analysis among patients with treatment-resistant depression

Mary L. Woody et al.

Summary: This study identified three RSFC-based subtypes in patients with treatment-resistant depression, which showed distinct aberrations in transdiagnostic cognitive-affective functioning across levels of analysis. These differences were maintained even after accounting for variability captured by disorder-specific depressive symptoms.

An fMRI study of cognitive reappraisal in major depressive disorder and borderline personality disorder

Victor De la Pena-Arteaga et al.

Summary: This study identified a shared neurobiological contributor to emotion regulation deficits in MDD and BPD characterized by decreased vlPFC activity, while also observing disorder-specific alterations. In MDD, there is a primary deficit in prefrontal activations, while BPD is defined by connectivity disruptions between the vlPFC and temporal emotion processing regions. These findings substantiate the different profiles of emotion regulation alterations observed in these disorders in neurobiological terms.
