The transport pathway in the ABCG2 protein and its regulation revealed by molecular dynamics simulations

Tamas Nagy et al.

Summary: Recent studies have provided atomic-level structural insight on the human ABCG2 membrane protein, shedding light on the complex dynamics of drug binding cavities and transport pathways. The findings suggest that membrane cholesterol can accelerate drug transport by promoting the closure of cytoplasmic protein regions, potentially influencing the pharmacokinetics of various clinically applied drugs.

Conformational changes in the nucleotide-binding domains of P-glycoprotein induced by ATP hydrolysis

Sepehr Dehghani-Ghahnaviyeh et al.

Summary: This study used molecular dynamics simulations to investigate the ATP hydrolysis-induced conformational changes in Pgp nucleotide-binding domains (NBDs), revealing that ATP/ADP exchange induced conformational changes within the conserved signature motif of NBDs, resulting in relative orientational changes. Furthermore, putative hydrolysis-competent configurations were captured, with a conserved glutamate in the Walker-B motif acting as a catalytic base capturing a water molecule likely initiating ATP hydrolysis.

Structure of the Human Cholesterol Transporter ABCG1

Liga Skarda et al.

Summary: ABCG1 is an ATP binding cassette transporter that removes excess cholesterol from peripheral tissues, and its mechanism of cholesterol transport is still unknown. Structural comparisons reveal differences in mechanism and substrate specificity between ABCG1, ABCG2, and ABCG5/G8, providing insights into ABCG1-mediated cholesterol transport.

Summary: In this study, three cryo-EM structures of human ABCG2 bound to anticancer drugs tariquidar, topotecan, and mitoxantrone were presented, revealing the binding pocket details and functional importance of key residues. Mutagenesis experiments and 3D variability analysis provided further insights into the mechanisms by which ABCG2 distinguishes between inhibitors and substrates.

Molecular basis of cholesterol efflux via ABCG subfamily transporters

Yingyuan Sun et al.

Summary: ABCG1 homodimer and ABCG5-ABCG8 heterodimer are crucial for maintaining cellular cholesterol levels. Cryo-EM structures reveal sterol-binding sites on both transporters and provide insights into the mechanisms of sterol recognition and export. Unique conformation of Walker A motif in G8 explains the asymmetry in ATPase activities of G5G8, offering a mechanistic framework for understanding cholesterol efflux via ABC transporters.

Structures of ABCG2 under turnover conditions reveal a key step in the drug transport mechanism

Summary: This study using cryo-EM structures of ABCG2 under turnover conditions reveals distinct conformational states, with two different turnover states observed. The relationship between ATP binding and closure of TMDs is highlighted as the likely bottleneck of the reaction cycle, where discrimination between substrates and inhibitors occurs.
