Polygenic basis and biomedical consequences of telomere length variation

Summary: This study characterized the genetic architecture of leukocyte telomere length in UK Biobank participants and identified 197 sentinel variants associated with LTL at 138 genomic loci. Genetically determined differences in LTL were found to be associated with various biological traits and diseases, and individuals with shorter LTL had a lower life expectancy compared to those with longer LTL. The findings provide insights into the genetic regulation of LTL, as well as its impact on physiological traits, diseases, and longevity.

Myofibrillar Structural Variability Underlies Contractile Function in Stem Cell-Derived Cardiomyocytes

Summary: This study aims to investigate the impact of myofibrillar structure on contractile function in iPSC-CMs. The results show that myofibrillar abundance is widely variable among iPSC-CMs and strongly correlates with contractile function. Controlling for myofibrillar structure can reduce false-positive findings, improve sensitivity for pharmacologic testing and disease modeling.

Summary: This study used human induced pluripotent stem cell-derived cardiomyocytes to reveal deficits in force generation, aberrant calcium handling, and increased reactive oxygen species levels in Duchenne muscular dystrophy (DMD) cardiomyocytes. The research found that progressive post-mitotic telomere shortening in DMD cardiomyocytes is associated with downregulation of shelterin complex and activation of the p53 DNA damage response.
