Non-linear Mendelian randomization analyses support a role for vitamin D deficiency in cardiovascular disease risk

Ang Zhou et al.

Summary: The study found a non-linear association between genetically predicted serum 25(OH)D concentration and CVD risk, with CVD risk initially decreasing steeply with increasing concentrations and leveling off at around 50 nmol/L. Correction of serum 25(OH)D level below 50 nmol/L was predicted to result in a 4.4% reduction in CVD incidence.

Summary: This study compared the prognostic values of different vitamin D biomarkers for total and cause-specific mortality in older adults and found that total, nonbioavailable, bioavailable, and free 25(OH)D were all inversely associated with mortality, with the strongest associations seen in participants with low vitamin D levels.

Vitamin D and Cancer: An Historical Overview of the Epidemiology and Mechanisms

Alberto Munoz et al.

Summary: This paper provides a narrative review of the evidence supporting the anticancer actions of vitamin D. It discusses the findings from ecological studies, observational studies, and case-control studies, as well as the limited success of meta-analyses of cancer incidence with respect to dietary intake. The review also highlights the effects of vitamin D compounds on tumor cell proliferation, dedifferentiation, invasion, and metastasis inhibition. Overall, the available mechanistic data support the global protective action of vitamin D against several important types of cancer.

Summary: In this study, treatment with vitamin D in patients with vitamin D deficiency and no prior history of myocardial infarction significantly reduced the risk of myocardial infarction and all-cause mortality. Maintaining vitamin D levels above 30 ng/mL was associated with a more significant reduction in the risk of myocardial infarction.

Summary: For patients with Vitamin D deficiency and no prior history of myocardial infarction, treatment to reach 25-hydroxyvitamin D levels above 20 ng/mL and 30 ng/mL is associated with significantly lower risk of all-cause mortality. The lower risk of myocardial infarction is only observed in individuals maintaining 25-OH levels above or equal to 30 ng/mL.
