Droplet digital PCR and mile-post analysis for the detection of F8 int1h inversions

Eric Manderstedt et al.

Summary: The study successfully designed and validated a sensitive and robust assay for detecting F8 Inv1 inversions, achieving high specificity and sensitivity. Milepost assays and droplet digital PCR were used to analyze DNA samples, showing concordance with known mutation status and estimating the detection limit of 2% for rare chromosome variants. The presented milepost analysis offers a sensitive and reliable method for detecting and quantifying F8 int1h inversions and other rearrangements involving intron 1.

Design of a digital-PCR assay to quantify fragmented human mitochondrial DNA

Alejandro Mosquera et al.

Summary: Digital PCR has been adapted for quantifying the proportion of mitochondrial DNA molecules with or without double-strand DNA breaks. The break-apart approach involves two differently labeled target sequences in the circular DNA molecule, where amplification in separate reaction partitions indicates fragmentation by DSBs. The technique has been validated using in vitro digestion with restriction endonucleases and shows potential for studying mtDNA in relation to aging, disease, and drug effects.
