ROS production and signalling in chloroplasts: cornerstones and evolving concepts

Christine H. Foyer et al.

Summary: ROS like singlet oxygen, superoxide, and hydrogen peroxide serve as markers of living cells and are produced abundantly during oxygenic photosynthesis. These ROS act as signals within chloroplasts, influencing cellular processes and redox signaling pathways. The challenge lies in understanding the organized delivery of regulated ROS from the photosynthetic electron transport chain to transmit redox signals from the environment to the nucleus, with stromal carbohydrate metabolism also playing a key role in chloroplast signaling pathways.

How does light facilitate vitamin C biosynthesis in leaves?

Takanori Maruta

Summary: Plants store high concentrations of ascorbate, especially in their leaves, which acts as an excellent antioxidant and photoprotectant. The d-mannose/l-galactose pathway is responsible for ascorbate biosynthesis in plants, which is facilitated by light intensity and requires photosynthesis. The activation of the rate-limiting enzyme GGP by light and photosynthesis plays a critical role in regulating ascorbate pool size. Ascorbate itself also regulates its biosynthesis by repressing GGP translation in a dose-dependent manner.

Activation of ascorbate metabolism by nitrogen starvation and its physiological impacts in Arabidopsis thaliana

Takumi Iwagami et al.

Summary: Redox homeostasis is crucial for plant acclimation to nutrient-deficient conditions. This study investigated the effects of nutrient deficiencies on antioxidant systems in Arabidopsis thaliana and found that nitrogen starvation increased ascorbate content. However, there were no phenotypic differences between wild type and ascorbate-deficient mutants under nitrogen starvation. Under high-light stress, the mutants experienced more severe photoinhibition than the wild type, but the same extent of photoinhibition was observed for both wild type and mutants when high-light stress and nitrogen starvation were combined.

Reactive oxygen species and organellar signaling

Su Yin Phua et al.

Summary: The evolution of photosynthesis in plants has been crucial but has also led to challenges in the form of reactive oxygen species (ROS) production. Multiple forms of ROS are generated in various plant cell compartments, requiring a sophisticated network of ROS detoxification and signaling tailored to individual organelles to safeguard the cell as a whole.

Distribution and Functions of Monodehydroascorbate Reductases in Plants: Comprehensive Reverse Genetic Analysis of Arabidopsis thaliana Enzymes

Mio Tanaka et al.

Summary: Monodehydroascorbate reductase (MDAR) is an enzyme involved in ascorbate recycling, with different numbers and distributions of isoforms in various plant species. The physiological significance of MDARs remains poorly understood. Classifying plant MDARs into three classes and studying Arabidopsis thaliana mutants lacking isoforms provide new insights into the roles of MDARs and their cooperation with other recycling systems.

Cooperation of chloroplast ascorbate peroxidases and proton gradient regulation 5 is critical for protecting Arabidopsis plants from photo-oxidative stress

Takashi Kameoka et al.

Summary: Under high-light stress, chloroplast APXs and pH-dependent mechanisms interact to maintain plants' stability against oxidative stress. The PGR5-dependent mechanisms compensate for the lack of APXs, and the cyclic electron flow dependent on the NADH dehydrogenase-like complex may play a role in regulating this process.

The d-mannose/l-galactose pathway is the dominant ascorbate biosynthetic route in the moss Physcomitrium patens

Tsubasa Sodeyama et al.

Summary: Ascorbate is an important redox compound in plants, primarily synthesized through the d-mannose/l-galactose pathway in streptophytes. Physcomitrium patens was found to possibly possess both the d-mannose/l-galactose pathway and the d-galacturonate pathway. Moss ALase was characterized as a functional enzyme involved in ascorbate metabolism, with PpALase1 playing a significant role in dehydroascorbate degradation rather than ascorbate biosynthesis.

The role of GDP-L-galactose phosphorylase in the control of ascorbate biosynthesis

Mario Fenech et al.

Summary: The study analyzed the subcellular localization and potential interactions of enzymes involved in L-ascorbate biosynthesis pathway in photosynthetic organisms. It was found that most enzymes showed dual cytosolic/nuclear localization and could interact with each other through coimmunoprecipitation, although direct interactions were not observed in yeast-two hybrid analysis. The research highlighted the crucial role of GDP-L-galactose phosphorylase (GGP) in regulating ascorbate synthesis and its impact on enzyme interactions within the pathway.
