Age-related matrix stiffening epigenetically regulates α-Klotho expression and compromises chondrocyte integrity

Summary: Matrix stiffening is a key characteristic of aged tissues. In this study, the authors found that an aged (stiff) matrix causes epigenetic repression of the gene encoding alpha-Klotho, a longevity factor, leading to chondrocyte dysfunction, which is a major cause of osteoarthritis. Exposing aged chondrocytes to a soft matrix restores a more youthful phenotype and enhances cartilage integrity. These findings highlight the importance of age-related biophysical alterations in extracellular matrix and have implications for the broader field of aging research.

Meta-analysis integrated with multi-omics data analysis to elucidate pathogenic mechanisms of age-related knee osteoarthritis in mice

Summary: This study used a combined approach of meta-analysis and multi-omics data analysis to uncover the pathogenic mechanisms of age-related knee osteoarthritis (KOA) in mice. Inflammation, autophagy, and cellular senescence were identified as primary hallmarks of age-related KOA. The Advanced Glycation End-Product (AGE)/Receptor for AGE pathway was found to be the most statistically robust pathway associated with age-related KOA. The molecular profiles of cartilage aging were distinct from those observed in surgically induced post-traumatic OA models, suggesting different mechanisms at play.

FSH directly regulates chondrocyte dedifferentiation and cartilage development

Summary: The study revealed that follicle-stimulating hormone (FSH) plays a critical role in chondrocyte dedifferentiation and cartilage destruction by decreasing type II collagen synthesis and inducing cytoskeletal reorganization. Knocking down the FSH receptor (Fshr) and inhibiting the Gai protein also affected chondrocyte dedifferentiation. Overall, high circulating FSH, independent of estrogen, is identified as an important regulator in chondrocyte dedifferentiation and cartilage destruction.
