Ring Opening of Epoxides by Pendant Silanols

Someshwar Nagamalla et al.

Summary: This study presents a new ring-opening reaction of epoxides catalyzed by pendant silanols. Different types of silanol epoxides derived from various allylic alcohols were compatible and yielded products with either endo- or exo-ring opening. An unusual rearrangement to tetrahydrofuran products was observed with one type of silanol epoxide derived from 4-alkenyl silanols. The utility of this methodology was demonstrated in a short preparation of protected D-arabitol.

Hydroxyselenylation and Tethered Silanoxyselenylation of Allylic Silanols

Harshit Joshi et al.

Summary: We present protocols for the highly regioselective hydroxyselenylation and silanoxyselenylation of allylic silanols. N-(Phenylseleno)phthalimide acts as the selenylating agent for both transformations. Hydroxyselenylation proceeds with >20:1 regioselectivity under basic conditions, and the products can be used as valuable synthons. The silanol plays a critical role in maintaining the yield and regioselectivity of this reaction. Surprisingly, under acidic conditions, only products of a tethered silanoxyselenylation are formed.

Summary: We demonstrate the effectiveness of di-tert-butylsilanols as nucleophiles for the interception of palladium pi-allyl species. The optimized protocol allows for stereospecific cyclization reactions, providing blank slates for further functionalization.

Ring Opening of Aziridines by Pendant Silanols Allows for Preparations of (±)-Clavaminol H, (±)-Des-Acetyl-Clavaminol H, (±)-Dihydrosphingosine, and (±)-N-Hexanoyldihydrosphingosine

Someshwar Nagamalla et al.

Summary: We present a unique strategy for the synthesis of vicinal amino alcohols through ring opening of aziridines with pendant silanols, which is compatible with a range of substrates. This method requires mild activation of the aziridine and tolerates various N-substituents. It has been successfully applied in the facile preparation of natural products and analogues.

Hexafluoroisopropyl Sulfamate: A Useful Reagent for the Synthesis of Sulfamates and Sulfamides

Matthew A. Sguazzin et al.

Summary: HFIPS is a bench-stable solid that reacts readily with a wide variety of alcohols, amines, phenols, and anilines under mild reaction conditions, producing hexafluoroisopropanol as the sole byproduct. The reaction products can often be isolated in high purity after an aqueous workup and removal of solvents by evaporation.

Tethered Silanoxyiodination of Alkenes

Ranjeet A. Dhokale et al.

Summary: This study presents the first examples of tethered silanoxyiodination reactions of allylic alcohols, yielding useful silanediol organoiodide synthons with high regioselectivity and diastereocontrol. The reaction is scalable without loss of yield or selectivity, and the products serve as starting materials for various transformations. DFT calculations provide insights into the excellent 6-endo selectivity of this reaction, indicating kinetically and thermodynamically favored products.

A Formal Rearrangement of Allylic Silanols

Ranjeet A. Dhokale et al.

Summary: The study demonstrates that 1M aqueous HCl/THF or NaBH4/DMF can facilitate the demercurative ring-opening of cyclic organomercurial synthons into secondary silanol products bearing terminal alkenes. A two-step protocol allows for the rearrangement of primary allylic silanol substrates into cyclic organomercurials. Computational investigations indicate that the rearrangement is thermodynamically controlled and the di-tert-butylsilanol protecting group plays a crucial role in product selectivity.

Diarylborinic Acid-Catalyzed Regioselective Ring Openings of Epoxy Alcohols with Pyrazoles, Imidazoles, Triazoles, and Other Nitrogen Heterocycles

Shrey P. Desai et al.

Summary: This study introduced a method for regioselective ring openings of 3,4- and 2,3-epoxy alcohols with ambident nitrogen heterocycles using a diarylborinic acid catalyst. It favors a single regioisomer in couplings of nucleophile and electrophile partners that exhibit low regioselectivity under conventional conditions, providing access to aromatic heterocycles with stereochemically defined, functionalized alkyl substituents similar in structure to medicinally relevant compounds.

Unusual rearrangement-remercuration reactions of allylic silanols

Someshwar Nagamalla et al.

Summary: The first examples of rearrangement reactions of allylic silanol substrates into linear ketone and 5-membered cyclic silanediol organomercurial products are presented in this study. The reactions are mediated by Hg(OTf)(2) and show differences in the use of base, solvent, and temperature. Mechanistic studies suggest that these products result from a series of cascading transformations in one pot, with a DFT analysis providing insights into the rearrangement process.
