Electrochemically driven cross-electrophile coupling of alkyl halides

Wen Zhang et al.

Summary: Recent research in medicinal chemistry has found a correlation between an increase in sp(3) carbons in drug candidates and their improved success rate in clinical trials. This study focuses on the development of robust and selective methods for constructing carbon(sp(3))-carbon(sp(3)) bonds. By using electrochemistry, the researchers achieve the selective activation of alkyl halides based on their electronic and steric properties, allowing for high selectivity in carbon(sp(3))-carbon(sp(3)) cross-electrophile coupling. This new protocol shows improved chemoselectivity and does not require a transition metal catalyst.

Nickel-Mediated Synthesis of Non-AnomericC-Acyl Glycosidesthrough Electron Donor-Acceptor Complex Photoactivation

Marcos Escolano et al.

Summary: This article presents a method for preparing nonanomeric C-acyl-saccharides from two different carboxylic acid feedstocks. The transformation is driven by the synergistic interaction of an electron donor-acceptor complex and Ni catalysis, enabling the coupling of redox-active esters onto preactivated pyranosyl- and furanosyl acids while maintaining their stereochemical integrity. The reaction occurs under mild conditions without the need for stoichiometric metal reductants or exogenous catalysts, using a commercially available Hantzsch ester as the organic photoreductant.

Accelerating reaction generality and mechanistic insight through additive mapping

Cesar N. Prieto Kullmer et al.

Summary: The generality of a reaction is crucial for determining its overall impact and usefulness in synthetic methods. This study developed an additive mapping approach that rapidly expands the utility of synthetic methods and provides mechanistic insight. By validating this approach on a specific reaction, the discovery of a phthalimide ligand additive was made, which overcomes limitations and has important mechanistic implications for nickel-catalyzed cross-couplings.

Synthesis of 2-Aryl Azetidines through Pd-Catalyzed Migration/Coupling of 3-Iodoazetidines and Aryl Boronic Acids

Han Yang et al.

Summary: A palladium-catalyzed cross-coupling reaction between 3-iodoazetidines and nonheteroaryl boronic acids was reported in this study. The reaction was facilitated by the [1,1'-biphenyl]-2-yldicyclohexylphosphane ligand, leading to the formation of 2-aryl azetidines. Control experiments indicated that the reaction can proceed through either a palladiumhydride/dihydroazete complex or free dihydroazete intermediate followed by hydropalladation.

Different Oxidative Addition Mechanisms for 12-and 14-Electron Palladium(0) Explain Ligand-Controlled Divergent Site Selectivity

Jacob P. Norman et al.

Summary: In cross-coupling reactions, dihaloheteroarenes typically react at C-halide bonds adjacent to a heteroatom. However, there have been anomalous reports where specific ligands cause inverted selectivity with dihalopyridines and -pyridazines. This study provides evidence that these ligands uniquely promote oxidative addition at 12e(-) Pd(0). Computations suggest that 12e(-) and 14e(-) Pd(0) can favor different mechanisms for oxidative addition due to differences in their HOMO symmetries, leading to different site preferences.

Arylboration of Enecarbamates for the Synthesis of Borylated Saturated N-Heterocycles

Grace L. Trammel et al.

Summary: Two catalytic systems have been developed for the arylboration of endocyclic enecarbamates, resulting in the synthesis of versatile borylated saturated N-heterocycles with good regio- and diastereoselectivities. The Cu/Pd dual catalytic reaction enables the synthesis of borylated, alpha-arylated azetidines, while the Ni-catalysed arylboration reaction efficiently functionalizes 5-, 6-, and 7-membered enecarbamates. The use of additives in the Cu/Pd system allows for a broader scope of reactions, and the products can be further manipulated to access biologically active compounds.
