Application of Raw and Chemically Modified Biomasses for Heterogeneous Cu-Catalysed Conversion of Aryl boronic Acids to Phenols Derivatives

Summary: This study investigates the ipso-hydroxylation of aryl boronic acids using cellulose and sugarcane bagasse as catalysts under environmentally friendly reaction conditions. The results show that CEDA-[Cu] material achieves the best catalytic efficiency under mild reaction conditions. The optimized catalytic sequence requires the presence of support material, Cu2O, and KOH at room temperature. The CEDA-[Cu] material is characterized and shows good recyclability after five catalytic cycles.

Phenols in Pharmaceuticals: Analysis of a Recurring Motif

Summary: Phenols and phenolic ethers are important molecular structures in nature and approved drugs. This study provides the first comprehensive compilation and analysis of the structures of U.S. FDA-approved molecules containing phenol or phenolic ether fragments. The research reveals the differences in physicochemical properties between phenols and phenol ethers, and their significance in drug development. Despite recent trends in increasing sp(3) centers in drug leads, phenols and phenolic ethers still have a significant presence in small-molecule drugs approved in 2020, indicating their special role in drugs and natural products.

A Method to Access Highly Functionalized Dibenzobicyclo[3.2.1]octadienones: Application to the Construction of the 6/6/5/6/6 Carbon Skeleton of Rubialatin A

Summary: The dibenzobicyclo[3.2.1]octadienone scaffold, found in various compounds such as naphthocyclinones, engelharquinones, and rubialatin A, has been successfully synthesized using a mild transition metal-free aryne insertion reaction. The methodology has been applied to the synthesis of the 6/6/5/6/6 scaffold of rubialatin A. H-1 NMR experimental studies confirm the reaction mechanism involves the formation of a benzocyclobutane followed by a 7-member carbocycle ring.

Generation of Fused Seven-Membered Polycyclic Systems via Ring Expansion and Application to the Total Synthesis of Sesquiterpenoids

Summary: Seven-membered polycyclic architectures, commonly found in natural products and molecular drugs, are challenging targets in synthesis. This study presents a base-induced ring expansion as a general strategy for constructing a wide range of fused seven-membered ring systems. The successful synthesis of two sesquiterpenoids, plecarpenene and plecarpenone, demonstrates the efficacy of this method for constructing fused bicyclo[5.3.0]decane skeletons.

Synthesis of Fused Pyrimido[1,6-a]indolones via Rhodium(III)-Catalyzed Cascade Annulations

Summary: A novel method for the synthesis of fused pyrimido[1,6-a]-indolone derivatives by annulation of 2-alkynylaryl aldehydes/2-alkynyl-arylidene ketones with N-(pivaloyloxy)-1H-indole-1-carboxamide catalyzed by rhodium has been successfully accomplished. The reaction proceeds through C-H activation based annulation with alkyne moiety followed by addition of nitrogen on to aldehyde/activated alkene to give the products in moderate to good yields. Highly fluorescent dipyrrinone analogues could be synthesized from the derived products.

Synthesis of γ-carboline N-oxide under gold(I)-catalysis and C-1 amino and fluoro γ-carboline

Summary: This study demonstrates the direct construction of the unknown gamma-carboline N-oxide scaffold at ambient temperature using intramolecular gold(I)-catalyzed cyclization of 2-alkynylindole-3-carbaldehyde oximes, with satisfactory to high yields of desired products. The scope and limitations of the methodology were evaluated, and the final compounds were fully characterized. Fluorination assays were also successfully achieved from the 3-phenyl-N-oxide derivative in two steps, resulting in the production of 1-fluoro-gamma-carboline.
