Discovery of novel drug-like antitubercular hits targeting the MEP pathway enzyme DXPS by strategic application of ligand-based virtual screening

Summary: In this manuscript, the authors successfully used ligand-based virtual screening (LBVS) to identify two small-molecule drug-like hit classes with excellent ADMET profiles against the challenging microbial enzyme DXPS. They introduced the concept of pseudo-inhibitors to overcome the limitations of standard computer-aided drug design tools for targets with large, hydrophilic binding sites. The hits displayed promising in vitro and whole-cell activity against M. tuberculosis DXPS, and possess unique mechanisms of inhibition, making them potential starting points for the development of antibiotics with a novel mode of action.

Thiamine analogues as inhibitors of pyruvate dehydrogenase and discovery of a thiamine analogue with non-thiamine related antiplasmodial activity

Summary: A series of derivatives of a triazole analogue of thiamine have been synthesized. The benzoyl ester derivatives were found to be potent thiamine pyrophosphate (TPP) competitive inhibitors and showed modest antiplasmodial activity, except for one derivative with low micromolar range activity. This activity was not affected by thiamine concentration or thiamine pyrophosphokinase expression.

Discovery of efficient inhibitors against pyruvate dehydrogenase complex component E1 with bactericidal activity using computer aided design

Summary: Computer aided optimization was utilized to design a series of new agrochemical compounds, which exhibited excellent inhibitory activity against Escherichia coli. Compound 6l showed the best inhibitory activity with an IC50 value of 95 nM. In vitro antibacterial activity tests showed that some compounds exhibited higher inhibition against Ralstonia solanacearum compared to commercial chemical pesticides.

Summary: OT and DAT are antimetabolites of thiamine. In this study, DATPP was found to be a stronger competitive inhibitor of PDHC compared to OTPP, but did not significantly impact cell growth in HeLa cell culture. The differences in effect on cell growth may be due to physicochemical properties and difficulty in DAT transport.
