Cp*Rh/Ag catalyzed C-H activation/cyclization sequences of NH-sulfoximines to fused aza-polyheterocycles under gentle conditions

Summary: A novel Rh/Ag co-catalyzed S-NH directed C-H activation and C-H/N-H bond functionalization protocol of free NH-sulfoximines with hypervalent iodonium ylides is disclosed. The C-H functionalization/cyclization sequences can be achieved at room temperature conditions with the aid of AgOTf. The reaction employs EtOH as a green solvent and only requires low catalyst loading under an oxygen/water-insensitive condition, allowing for the efficient preparation of a wide range of polyheterocyclic sulfoximines, including a complex pharmaceutical Folliculin analog.

The synthetic protocol showed broad functional group tolerance and mild reaction conditions, and can be viewed as the result of C-H activation, carbene insertion, and nucleophilic addition processes. The effectiveness of the synthesis protocol was supported by further product conversions and gram-scale reactions.

Rhodium-Catalyzed Enal Transfer with N-Methoxypyridazinium Salts

Summary: Herein, a simple method for functionalized enals involving enal-transfer reaction of water-soluble N-methoxypyridazinium salts is reported. This method proceeds under mild aqueous basic conditions through [2,3]-sigmatropic rearrangement of propargyl/allyl sulfur-ylides derived from in situ-generated Rh-(E)-enalcarbene. Various synthetically challenging allene- and allyl-functionalized (E)-enals with γ-C(sp(3)) quaternary center were obtained in good to high yields. Additionally, InCl3-catalyzed cascade cyclization of allenyl-enal and aniline resulted in a valuable pyrrolo[1,2-a]quinoline motif.

The gram-scale reaction and derivatization of the product demonstrate the synthetic practicality and utilization of this method.

Summary: A novel strategy for the synthesis of imidazo-fused polycyclic compounds under mild, base-free, and silver-free conditions has been reported. This step-economic protocol features the selective cleavage of two different C-H bonds in one step, and has the advantages of easy operation, readily available starting materials, gram-scale synthesis, broad functional group tolerance, and no requirement to presynthesize carbene precursors. The synthetic potential is demonstrated by the structural modification of a drug and the highly step-economic synthesis of Janus kinase inhibitor in only three steps.
