Dielectricity of a molecularly crowded solution accelerates NTP misincorporation during RNA-dependent RNA polymerization by T7 RNA polymerase

Summary: In biological systems, the synthesis of nucleic acids is catalyzed by enzymes in aqueous solutions. This study investigates the effects of polyethylene glycol on the fidelity of RNA synthesis and finds that it can promote the polymerization of non-complementary ribonucleotides. These findings suggest that changes in solution properties can alter the substrate specificity of RNA polymerase and increase genetic diversity.

Transcriptional Perturbations of 2,6-Diaminopurine and 2-Aminopurine

Summary: In this study, the effects of 2,6-diaminopurine (Z) on DNA transcription were systematically examined. The results showed that Z had an inhibitory effect on transcription without causing mutagenesis, while a structurally related adenine analog, 2-aminopurine (2AP), had no effect on transcription. Z was removed by transcription-coupled nucleotide-excision repair. These findings provide new insights into the role and biological functions of Z in transcription.

N1-methyl-pseudouridine is incorporated with higher fidelity than pseudouridine in synthetic RNAs

Summary: In vitro transcribed synthetic mRNAs are a new therapeutic modality. Uridine analogs are incorporated to enhance therapeutic efficacy and overcome immunogenicity. This study compared the fidelity of uridine analog incorporation using different RNA polymerases and found that m1 psi has higher fidelity than psi. The spectrum of mutations differs between RNA polymerases, but the distribution is not dependent on DNA sequence context.

Direct Nanopore Sequencing of Individual Full Length tRNA Strands
