ON-target Adverse Events of CRISPR-Cas9 Nuclease: More Chaotic than Expected

Off-target genotoxicity can be addressed with high fidelity Cas9 nucleases, while on-target genotoxicity is still underestimated. Structural basis of transposon end recognition explains central features of Tn7 transposition systems

Summary: The use of healthy donor T cells for CAR-T cell cancer therapy offers great potential, but the accuracy of gene editing tools and the requirement for T cell activation have limited their application. This study reports a high-fidelity RNA-guided endonuclease called Cas-CLOVER, which can achieve efficient multiplexed gene editing in resting T cells, combined with CAR transgene delivery to produce allogeneic CAR-T cells composed of high percentages of TSCM cells.

Functional Characterization of the N-Terminal Disordered Region of the piggyBac Transposase

Summary: The N-terminal disordered region (NTDR) of piggyBac DNA transposon plays an important role in the transposition process. Deletion of the NTDR significantly impairs piggyBac transposition, with varying effects in different cell types. The conservation of NTDR sequence is crucial for rescuing transposase activity, and the excision step of transposition is more severely affected by NTDR deletion. Bioinformatics analysis reveals that NTDR is specific for the piggyBac superfamily and is also present in transposase-derived proteins that are incapable of catalyzing transposition. These findings indicate the essential role of NTDR in fine-tuning transposition and its significance in the functions of piggyBac-originated co-opted genes.
