Summary: In a cuff model of peripheral neuropathy, endogenous acetylcholine continues to exert analgesic effects through nicotinic receptors, showing persistent efficacy in modulating nociceptive responses. Neuropathy does not affect the number or properties of dorsal horn cholinergic neurons, but rather alters the neurons receiving cholinergic signaling. Manipulating endogenous cholinergic signaling could be a potential avenue for treating mechanical allodynia in animal models of peripheral neuropathy.

Donepezil prevents morphine tolerance by regulating N-methyl-D-aspartate receptor, protein kinase C and CaM-dependent kinase II expression in rats

Summary: This study demonstrated that donepezil can attenuate morphine-induced tolerance by downregulating the expressions of NMDAR, PKC, and CaMKII pathways. Co-administration of donepezil with morphine prolonged the occurrence of analgesic tolerance in rats. Specifically, morphine upregulated NR1, PKC gamma, and CaMKII alpha in certain brain regions, while downregulating these proteins with donepezil treatment.
