Summary: The study shows that experience is not necessary for the development of synaptic morphology and the arrangement of different connections in local networks is stochastic. Loss of activity increases the variability in synaptic structure size, but does not disrupt their alignment. These results demonstrate that mechanisms for the structural diversity of neuronal synapses are intrinsic.

Summary: This study utilized a 3D-CLEM approach to reconstruct and analyze segments of basal dendrites of layer 2/3 PNs in the adult mouse somatosensory cortex, revealing that 10% of spines were dually innervated and 38% of inhibitory synapses localized to spines. The results suggest that spinous inhibition is more efficient than shaft inhibition, decoupling voltage and calcium signaling and potentially impacting synaptic plasticity.
