METTL3-mediated N6-methyladenosine modification governs pericyte dysfunction during diabetes-induced retinal vascular complication

Long Suo et al.

Summary: This study demonstrates the importance of m(6)A RNA modification in microvascular complications among patients with diabetes. METTL3, a regulator of m(6)A RNA methylation, is upregulated after diabetic stress and contributes to pericyte dysfunction through the regulation of PKC-eta, FAT4, and PDGFRA expression.

The m6A reader YTHDF2 is a negative regulator for dendrite development and maintenance of retinal ganglion cells

Fugui Niu et al.

Summary: In this study, the researchers found that the m(6)A reader YTHDF2 is highly expressed in mouse RGCs, and its knockout leads to increased dendrite branching and improved visual acuity. Furthermore, Ythdf2 knockout in the retina protects RGCs from dendrite degeneration caused by acute glaucoma. The study also identified the m(6)A-modified YTHDF2 target transcripts that mediate these effects, suggesting potential new treatment approaches for glaucoma.

Positive feedback regulation of microglial glucose metabolism by histone H4 lysine 12 lactylation in Alzheimer's disease

Rui-Yuan Pan et al.

Summary: The dysregulation of microglial function in Alzheimer's disease is associated with the glycolysis/H4K12la/PKM2 positive feedback loop. Inhibiting this loop can ameliorate pathological changes and cognitive deficits in a mouse model. This suggests that disrupting the positive feedback loop may be a potential therapeutic approach for treating AD.

ALKBH5-Mediated m6A Modification of A20 Regulates Microglia Polarization in Diabetic Retinopathy

Tingting Chen et al.

Summary: This study investigated the role of microglia polarization in the pathogenesis of diabetic retinopathy, as well as the mechanism of ALKBH5-mediated m(6)A modification of A20 in retinal microglia polarization. The results showed that diabetic rats had a higher M1 polarization rate and a lower M2 polarization rate of retinal microglia. With increasing glucose concentration, microglia tended to polarize into the M1 inflammatory type rather than the M2 anti-inflammatory type. Moreover, lower expression of A20 resulted in enhanced M1 polarization of retinal microglia in diabetic retinopathy, which was caused by ALKBH5-mediated m(6)A modification. This study may provide new perspectives on the pathogenesis, diagnosis, and treatment of diabetic retinopathy.
