Structural variants shape driver combinations and outcomes in pediatric high-grade glioma

Frank P. B. Dubois et al.

Summary: This study analyzed the contributions of structural variants (SVs) to the development of pediatric high-grade gliomas (pHGGs). The researchers found that the most recurrent SVs targeted MYC isoforms and receptor tyrosine kinases (RTKs), indicating an underappreciated role for MYC in pHGG. The study also identified different patterns of simple and complex SVs, with complex SVs associated with shorter overall survival in certain types of pHGGs. These findings provide important insights into the impact of SVs on gliomagenesis and tumor evolution.

Regional identity of human neural stem cells determines oncogenic responses to histone H3.3 mutants

Raul Bardini Bressan et al.

Summary: Point mutations within histone H3.3 are frequent in pediatric high-grade gliomas (pHGGs), with distinct mutations arising in different brain regions such as the forebrain and hindbrain. Regional differences in the response of fetal neural stem cells to these mutations suggest a possible role in tumorigenesis and tumor development.

Dissecting the impact of regional identity and the oncogenic role of human-specific NOTCH2NL in an hESC model of H3.3G34R-mutant glioma

Kosuke Funato et al.

Summary: The study reveals that the H3.3G34R-mutant glioma model recapitulates key features of the tumors, showing specificity to forebrain interneuronal progenitors; Mutations in H3.3G34R, ATRX, and TP53 cooperatively impact RNA splicing events and lead to increased expression of components of the Notch pathway, particularly NOTCH2NL gene family.

An atlas of cortical arealization identifies dynamic molecular signatures

Aparna Bhaduri et al.

Summary: Research using single-cell RNA sequencing and single-molecule fluorescent in situ hybridization techniques has revealed the early regional patterning process of the human brain. The study found differential gene expression in different areas early in the neocortex, with an increasing number of regional features as neural cells differentiate. The results demonstrate that early cortical areal patterning is influenced by strong gene expression markers, giving rise to multiple regions between the frontal and occipital poles.

Joint profiling of histone modifications and transcriptome in single cells from mouse brain

Chenxu Zhu et al.

Summary: Genome-wide profiling of histone modifications can provide insights into regulatory elements, cell-type-specific gene expression, and epigenetic regulatory mechanisms. An ultra-high-throughput method, Paired-Tag, allows joint profiling of histone modifications and transcriptome at single-cell resolution. Integrative analysis of the data reveals distinct groups of genes subject to divergent epigenetic regulatory mechanisms.

Brian Krug et al.

Summary: This review discusses the deregulation of the epigenome in primary brain tumors in children and young adults, highlighting recurrent mutations, protein expression, and epigenetic alterations that promote oncogenicity. The review also emphasizes the narrow developmental windows permitting oncogenic potential and the potential reversibility of altered chromatin states.

Widespread overexpression from the four DNA hypermethylated HOX clusters in aggressive (IDHwt) glioma is associated with H3K27me3 depletion and alternative promoter usage

Elisa Le Boiteux et al.

Summary: HOX gene deregulation may contribute to the development of adult glioma by leading to widespread upregulation of HOX genes, possibly through escape from DNA hypermethylation at CpG island promoters. The study provides a comprehensive description of epigenetic changes at HOX clusters and their contribution to transcriptional changes observed in adult glioma, as well as identifying potential 'master' HOX proteins that may contribute to the tumorigenic potential of glioma stem cells.

Single-cell atlas of early human brain development highlights heterogeneity of human neuroepithelial cells and early radial glia

Ugomma C. Eze et al.

Summary: Through single-cell RNA sequencing, nine progenitor populations at the earliest stages of human brain development were identified, including a highly prevalent mesenchymal-like population that disappears once neurogenesis begins. Comparison with mouse progenitor populations revealed two clusters enriched in the early stages of human cortical development. Organoid systems displayed low fidelity to neuroepithelial and early radial glia cell types, but improved as neurogenesis progressed.

A subset of pediatric-type thalamic gliomas share a distinct DNA methylation profile, H3K27me3 loss and frequent alteration of EGFR

Philipp Sievers et al.

Summary: A study identified a specific molecular class of pediatric-type malignant gliomas with EGFR amplifications or mutations, as well as H3.1 or H3.3 K27M mutations, all showing loss of H3K27me3 staining. While some tumors exhibited a bithalamic growth pattern, a significant proportion occurred in other locations.
