Hypoxia Reduction Sensitizes Refractory Cancers to Immunotherapy

Priyamvada Jayaprakash et al.

Summary: To support their growth, cancers need to increase their blood vessel density for oxygen and nutrient delivery. However, this irregular vasculature results in insufficient oxygen supply, leading to tumor hypoxia. In hypoxic and nutrient-deprived conditions, tumors and suppressive stroma thrive while antitumor immunity weakens. Reversing cancer hypoxia has the potential to improve the survival and function of tumor-infiltrating T cells and make tumors responsive to immunotherapy.

Immunogenic cell stress and death

Guido Kroemer et al.

Summary: Immunogenic cell death (ICD) plays an important role in therapy and disease, as dying mammalian cells release signals that interact with the host to determine the immunological correlates of cellular stress and death. ICD is crucial in immunosurveillance and is related to strategies evolved by pathogens and cancer cells. Additionally, normal cell death can also initiate antigen-specific immune responses.

Cytotoxic CD8+T cells in cancer and cancer immunotherapy

Hans Raskov et al.

Summary: The functions and interactions between the innate and adaptive immune systems play a crucial role in anticancer immunity. Cytotoxic T cells with CD8 as the most effective effectors in the anticancer immune response form the basis of successful cancer immunotherapies. Advancements in genetically modified or synthetic receptors on cytotoxic T cells are being developed for future cancer treatment. Combinatory regimens may optimize treatment effects and reduce adverse events in cancer therapy.

Mitochondrial stress induced by continuous stimulation under hypoxia rapidly drives T cell exhaustion

Nicole E. Scharping et al.

Summary: Continuous TCR signaling and hypoxia increase Blimp-1, suppressing mitochondrial reprogramming and generating reactive oxygen species, leading to T cell exhaustion. Mitigating T cell-intrinsic ROS and reducing tumor hypoxia can limit T cell exhaustion, synergizing with immunotherapy.
